We developed an all-optical method to measure the temperature on gold (nanorods and nanostars) magnetite nanoparticles under near-infrared radiofrequency excitation by monitoring excited state lifetime of Rhodamine B that lies within =/~20 nm from nanoparticle surface. reached high sensitivity (0.029 ± 0.001 ns/°C) low uncertainty (±0.3 °C). Gold nanostars are =/~3 =/~100 times more efficient than nanorods in inducing localized hyperthermia.
Adaptor protein 2 (AP2) is a major constituent of clathrin-coated pits (CCPs). Whether it essential for all forms clathrin-mediated endocytosis (CME) in mammalian cells an open issue. Here, we demonstrate, by live TIRF microscopy, the existence subclass relatively short-lived CCPs lacking AP2 under physiological, unperturbed conditions. This retained AP2-knockout and able to support internalization epidermal growth factor receptor (EGFR) but not transferrin (TfR). The AP2-independent...
Relatively few studies have examined the link between SNPs and mRNA translation, despite established importance of translational regulation in shaping cell phenotypes. We developed a pipeline analyzing allelic imbalance total polysome-bound mRNAs from paired RNA-seq data HCT116 cells identified 40 candidate tranSNPs, i.e. associated with allele-specific translation. Among them, SNP rs1053639 (T/A) on DNA damage-inducible transcript 4 (DDIT4) 3UTR was identified, reference T allele showing...
The teratogenic potential of commercially available copper oxide (CuO), titanium dioxide (TiO2) and zinc (ZnO) nanoparticles (NPs) was evaluated using the standardized FETAX test. After characterization NP suspensions by TEM, DLS AAS, histopathological screening advanced confocal energy-filtered electron microscopy techniques were used to characterize induced lesions track NPs in tissues. Except for nCuO, which found be weakly embryolethal only at highest concentration tested, did not cause...
Abstract Cyclin-dependent kinase 12 (CDK12) overexpression is implicated in breast cancer, but whether it has a primary or only cooperative tumorigenic role unclear. Here, we show that transgenic CDK12 the mouse mammary gland per se sufficient to drive emergence of multiple and multifocal tumors, while, cooperation with known oncogenes, promotes earlier tumor onset metastasis. Integrative transcriptomic, metabolomic functional data reveal hyperactivation serine-glycine-one-carbon network...
Asymmetric branched gold nanoparticles are obtained using for the first time in seed-growth approach a zwitterionic surfactant, laurylsulfobetaine, whose concentration growth solution allows to control both length base-width ratio of branches and LSPR position, that can be tuned 700–1100 nm near infrared range.
The biological players involved in angiogenesis are only partially defined. Here, we report that endothelial cells (ECs) express a novel isoform of the cell-surface adhesion molecule L1CAM, termed L1-ΔTM. splicing factor NOVA2, which binds directly to L1CAM pre-mRNA, is necessary and sufficient for skipping transmembrane domain ECs, leading release soluble latter exerts high angiogenic function through both autocrine paracrine activities. Mechanistically, L1-ΔTM-induced requires fibroblast...
Cancer stem cells (CSC) have been implicated in tumor progression. In ovarian carcinoma (OC), CSC drive formation, dissemination and recurrence, as well drug resistance, thus contributing to the high death-to-incidence ratio of this disease. However, molecular basis such a pathogenic role (OCSC) has elucidated only limited extent. context, functional contribution L1 cell adhesion molecule (L1CAM) OC stemness remains elusive. The expression L1CAM was investigated patient-derived OCSC. genetic...
The developmental toxicity of nanostructured materials, as well their impact on the biological barriers, represents a crucial aspect to be assessed in nanosafety policy framework. Nanosized metal oxides have been demonstrated affect Xenopus laevis embryonic development, with nZnO specifically targeting digestive system. To study mechanisms nZnO-induced intestinal lesions, we tested two different nominally sized ZnO nanoparticles (NPs) at effective concentrations. Advanced microscopy...
Under the action of near-infrared radiation, shape anisotropic gold nanoparticles emit two-photon luminescence and release heat. Accordingly, they have been proposed for imaging, photothermal therapies thermo-controlled drug delivery. In all these applications particular care must be given to control nanoparticle – cell interaction thermal efficiency nanoparticles, while minimizing their intrinsic cytotoxicity. We present here characterization newly developed branched nanostars, obtained by...
NUMB is a tumor suppressor gene that functions by inhibiting the action of NOTCH proto-oncogene and enhancing levels activity protein p53. In breast cancer (BC), loss-of-function (LOF), mediated various molecular mechanisms, frequent causal event. Herein, we establish loss protein, resulting from hyper-degradation, prevalent mechanism LOF in BC. Through RNAi-based screening, identified CRL7FBXW8 complex as E3 ligase responsible for hyper-degradation Genetic pharmacological inhibition rescued...
An unconventional endocytic regulatory process that directs TfR1 to lysosomal degradation in response iron is reported. This mechanism may represent a novel posttranslational control prevent overload.
Abstract The p140Cap adaptor protein is a tumor suppressor in breast cancer associated with favorable prognosis. Here we highlight function of orchestrating local and systemic tumor-extrinsic events that eventually result inhibition the polymorphonuclear myeloid-derived cell creating an immunosuppressive tumor-promoting environment primary tumor, premetastatic niches at distant sites. Integrative transcriptomic preclinical studies unravel controls epistatic axis where, through upstream...
Abstract The subversion of endocytic routes leads to malignant transformation and has been implicated in human cancers. However, there is scarce evidence for genetic alterations proteins as causative high incidence Here, we report that Epsin 3 (EPN3) an oncogene with prognostic therapeutic relevance breast cancer. Mechanistically, EPN3 drives tumorigenesis by increasing E-cadherin endocytosis, followed the activation a β-catenin/TCF4-dependent partial epithelial-to-mesenchymal transition...
Abstract One open question in the biology of growth factor receptors is how a quantitative input (i.e., ligand concentration) decoded by cell to produce specific response(s). Here, we show that an EGFR endocytic mechanism, non-clathrin endocytosis (NCE), which activated only at high concentrations and targets receptor degradation, requires tripartite organelle platform involving plasma membrane (PM), endoplasmic reticulum (ER) mitochondria. At these contact sites, EGFR-dependent,...
Advances in bladder cancer (BCa) treatment have been hampered by the lack of predictive biomarkers and targeted therapies. Here, we demonstrate that loss tumor suppressor NUMB promotes aggressive tumorigenesis worsens disease outcomes. Retrospective cohort studies show NUMB-loss correlates with poor prognosis post-cystectomy muscle-invasive BCa patients increased risk muscle invasion progression non-muscle invasive patients. In mouse models, Numb ablation induces spontaneous sensitizes...
