We investigated the effect of bone marrow cell (BMC) transplantation on established liver fibrosis. BMCs green fluorescent protein (GFP) mice were transplanted into 4-week carbon tetrachloride (CCl4)-treated C57BL6 through tail vein, and treated for 4 more weeks with CCl4 (total, 8 weeks). Sirius red GFP staining clearly indicated migrated existing along fibers, strong expression matrix metalloproteinase (MMP)-9 shown by anti-MMP-9 antibodies in situ hybridization. Double...

We here report nine liver cirrhosis (LC) patients that underwent autologous bone marrow cell infusion (ABMI) from the peripheral vein. Subjects were with LC total bilirubin of less than 3.0 mg/dl, platelet count more 5 (1010/l), and no viable hepatocellular carcinoma on diagnostic imaging. Autologous (BM; 400 ml) was isolated ilium under general anesthesia. Mononuclear cells (MNCs) separated by washing infused via MNC characteristics confirmed fluorescence‐activated sorting analysis (CD34,...

Recently, a new hepatitis B virus (HBV) mutant with HBe antigen-negative phenotype has been characterized, in which one TATA box-like motif of the precore/core promoter had degenerated: most frequently by both A-->T and G-->A mutations at positions 1762 1764, respectively. The clinical significance this is as yet unknown. In our present study, T1762 A1764 was sought sera from HBV-infected blood donors chronic liver disease patients directly sequencing PCR-amplified region HBVDNA. Also,...

Severe hypoglycemia causes neuronal death and cognitive impairment. Evidence suggests that hypoglycemic involves excitotoxicity DNA damage. Poly(ADP-ribose) polymerase-1 (PARP-1) normally functions in repair, but promotes cell when extensively activated by Cortical neuron cultures were subjected to glucose deprivation assess the role of PARP-1 death. -/- neurons wild-type, +/+ treated with PARP inhibitor 3,4-dihydro-5-[4-(1-piperidinyl)butoxy]-1(2H)-isoquinolinone both showed increased...

Apoptosis is implicated in neonatal hypoxic/ischemic (H/I) brain injury among various forms of cell death. Here we investigate whether overexpression heat shock protein (Hsp) 70, an antiapoptotic protein, protects the from H/I and pathways involved protection. Postnatal day 7 (P7) transgenic mice overexpressing rat Hsp70 (Tg) their wild-type littermates (Wt) underwent unilateral common carotid artery ligation followed by 30 mins exposure to 8% O(2). Significant neuroprotection was observed...

Hypoglycemia-induced brain injury is a significant obstacle to optimal blood glucose control in diabetic patients. Severe hypoglycemia triggers cascade of events vulnerable neurons that may culminate cell death even after normalization. A key event this the activation poly(ADP-ribose) polymerase-1 (PARP-1). Activated PARP-1 consumes cytosolic NAD, and because NAD required for glycolysis, hypoglycemia-induced render cells unable use when availability restored. Pyruvate, however, can be...

Abstract Glutathione (GSH) is a key antioxidant that plays an important neuroprotective role in the brain. Decreased GSH levels are associated with neurodegenerative diseases such as Parkinson’s disease and Alzheimer’s disease. Here we show diurnal fluctuation of correlated activity against oxidative stress dopaminergic cells. In addition, found cysteine transporter excitatory amino acid carrier 1 (EAAC1), which involved neuronal synthesis, negatively regulated by microRNA miR-96-5p,...

Peroxynitrite can nitrate tyrosine residues of proteins. We examined nitrotyrosine-containing proteins in cerebrospinal fluid 66 patients with neurogenic disease by immunoblot analysis. Nitrated residue-containing protein was observed the and concluded to be manganese superoxide dismutase (Mn-SOD). The nitrated Mn-SOD level strikingly elevated amyotrophic lateral sclerosis slightly increased Alzheimer's Parkinson's patients, whereas an only progressive supranuclear palsy group.

Abstract Excitatory amino acid carrier 1 (EAAC1) is a glutamate transporter expressed on mature neurons in the CNS, and primary route for uptake of neuronal cysteine needed to produce glutathione (GSH). Parkinson's disease (PD) neurodegenerative disorder pathogenically related oxidative stress shows GSH depletion substantia nigra (SN). Herein, we report that 1‐methyl‐4‐phenyl‐1,2,3,6‐tetrahydropyridine (MPTP)‐treated mice, an experimental model PD, showed reduced motor activity, contents,...
