Abstract Although long noncoding RNAs (lncRNAs) dominate the transcriptome, their functions are largely unexplored. The extensive overlap of lncRNAs with coding and regulatory sequences restricts systematic interrogation by DNA-directed perturbation. Here we developed genome-scale lncRNA transcriptome screening using Cas13d/CasRx. We show that RNA targeting overcomes limitations inherent to other methods, thereby considerably expanding explorable space lncRNAome. By evolving system toward...

In using Western blot analysis with antibodies raised against recombinant pyruvate dehydrogenase kinase (PDK) isoforms PDK2 and PDK4, this study demonstrates selective PDK isoform switching in specific skeletal muscle types response to high-fat feeding that is associated altered regulation of activity by pyruvate. The administration a diet high saturated fats led stable (approximately 2-fold) increases activities both typical slow-twitch (soleus [SOL]) fast-twitch (anterior tibialis [AT])...

AMPK activates SIRT1 in liver and skeletal muscle. Impaired circadian function is associated with development of obesity. regulates suppressed white adipose tissue (WAT) obese patients. We examined the potential role regulation components WAT db/db mice fed a high-fat diet (HFD), investigated whether metformin-mediated activation opposed any deleterious changes clock mechanism.db/+ were administered metformin (250 mg/kg/day; 7 days). Separately, HFD for 16-weeks. 3T3-L1 adipocytes incubated...

Rapid inhibition of acetyl-CoA carboxylase (ACC) activity in rat liver response to 6 h starvation and rapid re-activation 2-6 re-feeding chow were shown be due changes the expressed existing enzyme. Decreases increases ACC concentration occurred at later stages transitions, i.e. 6-48 8-24 respectively. The decrease was primarily its phosphorylation state, demonstrated by a significantly decreased Vmax. increased Ka for citrate enzyme purified avidin-Sepharose chromatography from h- or 48...

Solid cancers like pancreatic cancer (PDAC) frequently exploit nerves for rapid dissemination. This neural invasion (NI) is an independent prognostic factor in PDAC, but insufficiently modelled genetically-engineered mouse models (GEMM) of PDAC. Here, we systematically screened human-like NI Europe's largest repository GEMM PDAC comprising 295 different genotypes. phenotype screen uncovered two with NI, which are both characterized by pancreas-specific overexpression...

A comparison was made between the time courses of restoration pyruvate dehydrogenase activities, fructose 2,6-bisphosphate concentrations and lipogenic rates, together with net hepatic glucose flux glycogen synthesis/deposition in livers 48 h-starved rats provided laboratory chow ad libitum for up to 24 h. Increased glycogenesis, lipogenesis uptake were observed after 1 h re-feeding, preceding re-activation dehydrogenase, which occurred 3-4 only 5-6 The implication temporal relationship...

We investigated the temporal relationship between hepatic glycogen depletion and cardiac PDH (pyruvate dehydrogenase complex) activities during acute phase of starvation. There was a striking correlation decline in inactivation first 10 h Re-feeding after 6 starvation associated with complete re-activation liver to approx. 75% fed value heart, whereas rats previously starved for 24-48 delayed diminished heart. The results are discussed reference fate dietary carbohydrate re-feeding.

The progressive effects of starvation on muscle glucose utilization were studied in the conscious resting rat. High rates uptake and phosphorylation constantly working cardiothoracic (heart, diaphragm) postural skeletal muscles (soleus, adductor longus) maintained for at least 9 h starvation. A rapid decline cardiac was observed during period 9-24 starvation, but other more gradual. Consequently, even after 24 h, these remained quantitatively significant. In both (postural) muscle, activity...

The mitochondrial pyruvate dehydrogenase complex (PDC) catalyses the oxidative decarboxylation of pyruvate, and links glycolysis to tricarboxylic acid cycle ATP production. Adequate flux through PDC is important in tissues with a high requirement, lipogenic (since it provides cytosolic acetyl-CoA for fatty (FA) synthesis), generating malonyl-CoA, potent inhibitor carnitine palmitoyltransferase (CPT I). Conversely, suppression activity crucial glucose conservation when scarce. This review...

Major decreases PDHa activities all these were observed after 15 h starvation, by which time less than 40% values. general, response starvation was more rapidly high oxidative capacity.
