Immune checkpoint blockade (ICB) therapy potently revives T cell's response to cancer. However, patients suffered with tumors that had inadequate infiltrated immune cells only receive limited therapeutic benefits from ICB therapy. Synthetic biology promotes the alternative strategy of harnessing tumor-targeting bacteria synthesize therapeutics modulate immunity in situ. Herein, we engineered attenuated Salmonella typhimurium VNP20009 gene circuits synthetize granulocyte-macrophage...

Dendritic cells (DCs) can form a T cell niche in tumor microenvironment and drive proliferation through immunostimulatory ligands. However, hypoxia severely hinders the infiltration of immune cells, thus restricting cells' anti-tumor response. Microalgae have evolved an efficient photosynthesis system that might be promising oxygen source to mitigate hypoxia. Here, we combine Chlorella vulgaris genetically engineered DC membrane, overexpressing necrosis factor (TNF) ligand proteins OX40L,...

An important factor in the development of type 1 diabetes (T1D) is deficiency inhibitory immune checkpoint ligands, specifically programmed cell death ligand (PD-L1) and galectin-9 (Gal-9), β-cells. Therefore, modulation pancreas-infiltrated T lymphocytes by exogenous PD-L1 or Gal-9 an ideal approach for treating new-onset T1D. We genetically engineered macrophage cells to generate artificial extracellular vesicles (aEVs) overexpressing Gal-9, which could restrict islet autoreactive protect...

Inactivated tumor cells preserve a whole spectrum of antigens, including neoantigens that can elicit potent immune response. Nevertheless, avoiding tumorigenic risks while stimulating robust anti-tumor response remains pivotal challenge. Herein, we prepare enucleated cancer decorated with bacteria-derived vesicles (BDVs) carrying stimulator interferon gene (STING) agonists as live cell vaccines. Enucleated retain intact morphology excepting the nucleus. BDVs loaded STING agonist cyclic...

How does the brain support 'wellbeing'? Because it is a multidimensional construct, likely product of multiple co-active networks that vary across individuals. This perhaps why prior neuroimaging studies have found inconsistent anatomical associations with wellbeing. Furthermore, these used 'laboratory-style' or 'resting-state' methods not amenable to finding manifold networks. To address issues, we had participants watch full-length romantic comedy-drama film during functional magnetic...

Autoimmune destruction of pancreatic β-cells leads to impaired insulin production and onset type 1 diabetes (T1D). Hence, immunomodulation pancreas-infiltrated immune cells especially the autoreactive-T is a promising way hinder reverse progress T1D. The process of complex cognition, which includes language processing, is dynamic in nature and involves various network modes or cognitive modes. This can be manifested by a set brain states transitions between them. Previous neuroimaging studies have shed light on how bilingual brains support native (L1) second (L2) through shared network. However, the mechanism this enables L1 L2 processing remains unknown. study examined issue testing hypothesis that associated with distinct state...
