Simple methods have been developed for encapsulating aqueous solutions of protein within polymer membranes. Stable microcapsules 1 to 100 μ in diameter, with semipermeable membranes, can be made by depositing around emulsified droplets, either interfacial coacervation or polycondensation. Aqueous suspensions enzyme-loaded act well on small-molecular substrates both vitro and vivo.
Phenylketonuria (PKU), with its associated hyperphenylalaninemia (HPA) and mental retardation, is a classic genetic disease the first to have an identified chemical cause of impaired cognitive development. Treatment from birth low phenylalanine diet largely prevents deviant phenotype by ameliorating HPA recognized as one effective treatments disease. However, compliance dietary treatment difficult when it for life, now recommended internationally used set guidelines, probably unrealistic....
Suspension of living hepatocytes were microencapsulated inside 300 micron mean diameter alginate artificial cells. The galactosamine fulminant hepatic failure rat model was used. 48 hours after the injection galactosamine, grade II coma rats divided into pairs. One pair randomly chosen for control group, and other treated group. Each in group received one peritoneal microcapsules containing no hepatocytes. survival is significantly higher than
Control trials and statistical analysis were carried out to assess the effects of albumin-collodion microencapsulated activated charcoal (ACAC) hemoperfusion on fulminant hepatic coma. A rat model galactosamine induced coma was used. Rats which did not recover died at 3.0+/-0.6 days after injection. Those survived this period recovered. Forty-eight hours injection, a test group 21 rats treated with 1 hour compared an untreated 23 rats. 71.4% as 30.4% Statistical (t test) shows significant...
We crosslink hemoglobin (Hb), superoxide dismutase (SOD), catalase (CAT), and carbonic anhydrase (CA) to form a soluble polyHb-SOD-CAT-CA nanobiotechnological complex. The obtained product is complex with three enhanced red blood cell (RBC) functions without group antigens. In the present study, 2/3 of volume was removed result in 90-min hemorrhagic shock at mean arterial pressure (MAP) 30 mmHg. This followed by reinfusion different resuscitation fluids, then for another 60 min....
The mitochondrial Ca2+ uniporter is the channel responsible for uptake. It plays crucial physiological roles in regulating oxidative phosphorylation, intracellular signaling, and cell death. contains pore-forming MCU subunit, auxiliary EMRE protein, regulatory MICU1 which blocks pore under resting cellular concentrations. has been known decades that spermine, a biological polyamine ubiquitously present animal cells, can enhance uptake, but underlying mechanisms remain incompletely...
In strokes, myocardial infarctions, severe sustained hemorrhagic shock, and donor organs, inadequate blood supply results in lack of oxygen to the tissue (ischemia). If ischemia is sustained, reperfusion with needed can result injury (ischemia-reperfusion injury) due formation reactive species. We are studying an oxygen-carrying solution anitoxidant activity formed by cross-linking hemoglobin, superoxide dismutase, catalase form PolyHb-SOD-CAT. The present report studies its effect on...
We have recently reported our study on novel nano‐dimension red blood cell (rbc) substitute based ultrathin PEG‐PLA membrane nanocapsules (80–150 nanometer diameter) containing hemoglobin (Hb) and enzymes. These a markedly increased the circulation half‐times as compared to earlier PLA nanocapsules. In present be here, instead of looking at this from pharmacodynamic point view, we design Hb view transfusion medicine. For instance, maximal levels systemic non‐red that can attained after one...
Polyhemoglobin-superoxide dismutase-catalase-carbonic anhydrase (PolyHb-SOD-CAT-CA) is a therapeutic antioxidant that also transports both oxygen and carbon dioxide. This formed by crosslinking Hb with SOD, CAT, CA using glutaraldehyde. Crosslinking stroma free from red blood cell (rbc) reduces activity to 55%. Addition of more resulted in preparation the same as RBC. PolyHb complex acts buffer prevent large pH changes dioxide converted carbonic acid. We then prepare optimize novel PolyHb-SOD-CAT-CA,
Under specific conditions, bone marrow cells can transdifferentiate into a variety of cell types including hepatocytes. In this study, bioencapsulated were transplanted intraperitoneally 90% hepatectomized rats. We then followed the transdifferentiation and effect on liver regeneration in failure model. Bone isolated from Wistar rats using alginate-polylysine-alginate method. These Blood chemistry, HGF, weight, survival recipient evaluated. Histology immunocytochemistry used to analyze...
