[A transcriptomic study of osteoporosis induced by ketogenic diet in mice].

ABSTRACT
To investigate the molecular mechanism of osteoporosis caused by ketogenic diet (KD) using transcriptomic analysis.Sixteen 8-week-old female C57BL/6J mice were divided into KD group and sham for feeding with normal 3 months, respectively. Body weight, blood glucose ketone levels measured every two weeks. Microstructure cancellous bone in distal femur was observed Micro-CT. Total RNA extracted from marrow cells analysis bioinformatics analysis. RT-qPCR used to verify expression genes significant differential between groups.KD obviously weakened microstructure mice. Compared those group, showed 165 differentially expressed (94 up-regulated 71 down-regulated ones), including Acot1, Mpig6b, Gp9, Ppbp, Slc2a9, etc. KEGG pathway enrichment obvious Apelin signaling pathway, PI3K- Akt ECM-receptor interaction signal transduction greater number genes. RTqPCR results that 5 screened transcriptomics significantly upregulated among which Mpig6b Ppbp over folds (2.49 ± 0.665, 2.58 0.470, 2.59 0.611, respectively), suggesting their involvement KD-induced osteoporosis.The enriched pathways identified mouse models provide new clues studying prevention osteoporosis.
