Prolongation of mouse heart allograft survival by injecting T.gondii soluble tachyzoite antigen

ABSTRACT
Objective To investigate the effects of T. gondii soluble tachyzoite antigen (STAgs) on survival time mouse heart allograft and possible mechanism. Methods The STAgs were prepared by pulverizing with ultrasound ice. Cervical heterotopic transplantations done using Balb/c mice as donors, C57BL/6 recipients.The recipients classified randomly into three groups: syngeneic group, acute rejection group STAgs-treated group. in injected subcutaneously 0. 1 ml PBS (5 μg) at 4th day before transplantation respectively, those not subjected to any treatment. grafts observed daily cervical palpation, total cessation cardiac contraction was defined endpoint. allografts harvested 7th after for pathological examination immunohistochemical staining CD4+ T, CD8+ Results all alive 100th transplantation. average (6.7± 0.5) days (70.8± 3.5) days,respectively (P<0.05). HE showed that fallen 0 degree, Ⅲ-Ⅳ degree 0- Ⅰ respectively. Immunohistochemical revealed T CD8+T markedly down-regulated compared Conclusion can significantly prolong inhibit probably changing ratio TH1/TH2, or inhibiting effect dendritic cells inducing lipoxin A4.
