ABSTRACT
Objective To investigate the expression of inducible nitric oxide synthase (iNOS) and levels (NO) in THP-1 J774A.1 cells during Leptospira interrogans (L.interrogans) infection for a better understanding mechanism macrophages involved defense against L. strains different hosts. Methods The human mononuclear (THP-1) murine (J774A.1) were infected with strain 56601. The iNOS at mRNA protein determined by using real-time RT-PCR flow cytometry analysis. NO detected Griess test. Results The level 2, 4, 12 24 hours respectively 1.37, 2.82, 25.76, 27.47 times 1.59, 3.98, 3.89, 8.81 than that without (P<0.05). rates increased from 34.16% to 85.85%, 93.82%, 91.77% 93.65% along time up-regulated 22.08% 72.64%, 81.33%, 80.03% 65.72% after (P<0.05), respectively. Results test indicated 0.1588 μmol/L 0.2208 μmol/L, 0.2668 0.3808 0.3828 0.0988 0.2848 0.3228 0.2608 0.3308 (P<0.05). Conclusion The significantly infection.
