ABSTRACT
Objective To measure the expression of protein kinase D1 (PKD1) , tyr463-phos-phorylaed PKD1 (pPKD1-tyr463) and ser916-phos-phorylaed (pPKD1-ser916) in squamous cell carcinoma (SCC) Bowen's disease (BD) actinic keratosis (AK) to explore their significance. Methods Fresh tissue samples were resected from lesions patients with SCC (SCC group) BD (BD AK (AK as well normal skin healthy human controls (control each group had a sample size 10. Real-time RT-PCR was performed mRNA gene (PRKD1) Western blot analysis determine PKD1, pPKD1-tyr463 pPKD1-ser916. In addition, immunohistochemical study conducted pPKD1-ser916 another 50 paraffin-embedded SCC, 20 BD, 10 samples. Results PRKD1 significantly differed among control (0.64 ± 0.09) (5.37 1.06) (2.69 0.72) (2.43 0.46) (F= 21.37, P 0.05) . Immunohistochemical showed that total groups mainly expressed cytoplasm membrane spinous layer cells atypical cells, rates higher than those (all < 0.01) The only slightly some cancer nests well-differentiated tissues, but not poorly-differentiated AK, tissues tissues. group, rate increased increase pathological grade positively correlated (rc c= 0.479, results consistent findings. Conclusion PKD1 may be involved development differentiation tumors derived stratified epithelium, exert promotive effects on formation cutaneous by activating Tyr463 phosphorylation site.
