Effect of 5,6-dihydrocyclopenta-1, 2-dithiole-3-thione on Müller cells under the high glucose

ABSTRACT
Objective
To investigate the cellular viability and mitochondrial reactive oxygen species (ROS) production of Muller cells under high glucose condition, explore protection role 5,6-dihydrocyclopenta-1, 2-dithiole-3-thione (CPDT) on cells.
Methods
Muller from Sprague Dawley rats were divided into 5 groups randomly, including 25 mmol/L normal group (group A) 65 B). High with 45, 60, 70 μmol/L CPDT cultured them 72 hour was set as C, D E. Water soluble tetrazolium salt (WST)-8 used to measure viability. Flow cytometry active apoptosis index. The expression nuclear factor erythroid 2-related 2 (Nrf2), hemeoxygenase-1 (HO-1), Bcl-2 Bax protein measured by Western blot.
Results
Compared A, WST-8 showed that apparently decreased in B (t=39.59, P 0.05), but recovered E (t=-4.17, -7.52; P<0.05). Compared FCM ROS levels higher (t=-30.99, B, (t=27.68, Bax, Nrf2 HO-1 increased (t=-11.03, –63.17, –11.44; P<0.05), while bcl-2 (t=7.861, Nrf2, (t=15.11, 26.59, 6.27; (t=-6.53, P<0.05).
Conclusions
Under glucose, may reduce cells. It inhibit through activating Nrf2/HO-1 pathway balancing level ROS.
