Effect of Shen fu injection on the expressions of ORP150, XBP1 and CHOP mRNAs in hypoxic-ischemic neonatal rats

ABSTRACT
Objective
To investigate the effect of Shenfu injection on expressions endoplasmic reticulum stress-related factors oxygen-regulated protein 150 (ORP150), X-box binding 1(XBP1), and C/EBP homologous (CHOP) mRNAs in cerebral cortical neurons after hypoxic-ischemic brain damage (HIBD) neonatal rats.
Methods
The 7 day-newborn Sprague-Dawley rats were randomly divided into sham operation, normal saline treatment (Shenfu injection, 10ml/kg per day, peritoneal for 3 days) groups. They redivided 3h, 6h, 12h, 24h, 3d 7d subgroups at different time pointsafter modeling(n=8 each group). A HIBD model was induced. Reverse transcriptionpolymerase chain reaction used to detect ORP150, XBP1, CHOP rat cortex.
Results
The XBP1 operation group very week. The significantly upregulated compared those 3h modeling (all P< 0.05); reached peak 6 respectively. Then they decreased mildly closed level day 7. 12, 24h lower than 0.05). mRNA expression positively correlated with (r=0.649, 0.05).
Conclusions
HIBD induces stress response. may be involved delayed neuronal process rats. downregulates inhibits response.
