Understanding how the extracellular matrix impacts function of cancer stem cells (CSCs) is a significant but poorly understood problem. We report that breast CSCs produce laminin (LM) 511 promotes self-renewal and tumor initiation by engaging α6Bβ1 integrin activating Hippo transducer TAZ. Although TAZ important for CSCs, mechanism unknown. observed regulates transcription α5 subunit LM511 formation matrix. These data establish positive feedback loop involving contributes to stemness in cancer.

Although the α6β1 integrin has been implicated in function of breast and other cancer stem cells (CSCs), little is known about its regulation relationship to mechanisms involved genesis CSCs. We report that a CD44high/CD24low population, enriched for CSCs, comprised distinct epithelial mesenchymal populations differ expression two α6 cytoplasmic domain splice variants: α6A α6B. α6Bβ1 defines population necessary CSC function, cannot be executed by integrins. The generation tightly controlled...

We show that the VEGF receptor neuropilin-2 (NRP2) is associated with high-grade, PTEN-null prostate cancer and its expression in tumor cells induced by PTEN loss as a consequence of c-Jun activation. VEGF/NRP2 signaling represses insulin-like growth factor-1 (IGF-IR) signaling, mechanism involves Bmi-1-mediated transcriptional repression IGF-IR. This has significant functional therapeutic implications were evaluated. IGF-IR positively correlates inversely NRP2 tumors. robust biomarker for...

Abstract Extracellular heat shock protein-90alpha (eHsp90α) plays an essential role in tumour invasion and metastasis. The plasma eHsp90α levels patients with various cancers correlate the stages of diseases. Nonetheless, mechanism action by tumour-secreted remained unclear. Here we show that accounts for approximately 1% total cellular Hsp90α is associated exosomes. CRISPR-cas9 knockout did not affect overall distribution quantity secreted exosomes, but it caused increased...

Alzheimer's disease (AD) is the leading cause of dementia globally, but effective treatment lacking. We aimed to explore lncRNA XIST role in AD and mechanisms involved effect changes on expression Aβ-degrading enzymes. The mouse model cell induced by Aβ were established. LncRNA XIST, IDE, NEP, Plasmin, ACE, EZH2 expressions distribution nucleus cytoplasm detected qRT-PCR. Inflammatory cytokines IL-6, IL-1β, TNFα, IL-8, Aβ42 levels ELISA. Long noncoding RNAs (lncRNAs) have been confirmed to be aberrantly expressed in various diseases including tumors. Recently, a new tumor-related lncRNA, lncRNA TRPM2 antisense RNA (TRPM2-AS), was shown involved many tumors, such as lung cancer and breast cancer. However, the expression role of TRPM2-AS development gastric (GC) not elucidated. In current study, we provided evidence that levels were increased both GC tissues cell lines. We also showed overexpression modulated by ELK1,...

Estrogen receptor β (ERβ) promotes the degradation of hypoxia inducible factor 1α (HIF-1α), which contributes to ability this hormone sustain differentiation epithelial and carcinoma cells. Although loss ERβ consequent HIF-1 activation occur in prostate cancer with profound consequences, mechanism by HIF-1α is unknown.
