Abstract This study was designed to develop a bioactive scaffold enhance bone defect repair in steroid‐associated osteonecrosis (SAON). Icaritin, metabolite of the herb Epimedium , has been identified as an angiogenic and osteogenic phytomolecule. Icaritin homogenized into poly lactic‐ co ‐glycolic acid/tricalcium phosphate (PLGA/TCP) form icaritin‐releasing porous composite (PLGA/TCP/icaritin) by fine‐spinning technology. In vitro, high performance liquid chromatography used determine...
Abstract Background Sox6 is a multi-faceted transcription factor involved in the terminal differentiation of many different cell types vertebrates. It has been suggested that mice as well zebrafish plays role skeletal muscle by suppressing slow fiber specific genes. In order to understand how coordinately regulates multiple type genes during development, we have performed ChIP-seq analyses identify target mouse fetal myotubes and generated muscle-specific knockout (KO) determine null...
We found that Icaritin, an intestinal metabolite of Epimedium-derived flavonoids (EF) enhanced osteoblastic differentiation mesenchymal stem cells (MSCs) only under osteogenic induction conditions. also demonstrated its effect on inhibition adipogenic MSCs. Unlike the findings others EF compounds, we showed Icaritin was unable to promote proliferation, migration and tube like structure formation by human umbilical vein endothelial (HUVECs) in vitro. These results suggested exogenous...
Nondioxin-like polychlorinated biphenyls (NDL PCBs) activate ryanodine-sensitive Ca 2þ channels (RyRs) and this activation has been associated with neurotoxicity in exposed animals.RyR-active congeners follow a distinct structure-activity relationship quantitative (QSAR) predicts that large number of PCBs likely the receptor, which requires validation.Additionally, previous structural based conclusions have established using receptor ligand binding assays but impact varying PCB structures on...
Nineteen ortho-substituted PCBs are chiral and found enantioselectively enriched in ecosystems. Their differential actions on biological targets not understood. PCB 95 (2,2′,3,5′,6-pentachlorobiphenyl), a of current environmental relevance, is among the most potent toward modifying ryanodine receptors (RyR) function Ca2+ signaling. enantiomers separated assigned aR- aS-PCB using three chiral-column HPLC circular dichroism spectroscopy. Studies RyR1-enriched microsomes show aR-PCB with >4×...
Non–dioxin like polychlorinated biphenyls (NDL-PCBs) are legacy environmental contaminants with contemporary unintentional sources. NDL-PCBs interact ryanodine receptors (RyRs), Ca2+ channels of sarcoplasmic/endoplasmic reticulum (SR/ER) that regulate excitation-contraction coupling (ECC) and Ca2+-dependent cell signaling in muscle. Activities 4 chiral congeners PCB91, 95, 132, 149 their respective 4- 5-hydroxy (-OH) derivatives toward rabbit skeletal muscle receptor (RyR1) investigated...
