We have analyzed the functions encoded by bgl operon in Escherichia coli K-12. Based on ability of cloned regions to complement a series Bgl- point mutations, we show that three structural genes, bglC, bglS, and bglB, are located downstream regulatory locus bglR order indicated. Using bgl-lacZ transcriptional fusion, bglC bglS involved regulating expression. The presence gene trans is absolutely required for expression which constitutive when only present. When genes both present cell,...
Abstract Sialidase is shown to be localized in the lysosomes of rat liver and kidney. The pH optimum with sialyllactose as substrate 4.4, glycoproteins 4.0. enzyme not affected by many divalent metal ions, ethylenediaminetetraacetate, or p-chloromercuribenzoate, active against gangliosides, indicating its possible role their intracellular digestion.
Abstract A variety of rat tissues have the ability to hydrolyze 1-l-β-aspartamido[-2-acetamido]-1,2-dideoxy-β-d-glucose. In liver and kidney enzyme is present in lysosomes. The lysosomal nature was established by amount sedimentable with mitochondrial light particles, purification lysosomes, isolation Triton WR 1339-filled Freezing thawing, osmotic shock, treatment X-100 resulted release from purified 430-fold Sephadex gel filtration 650-fold chromatography on carboxymethyl cellulose. both...
Mixed rumen micro-organisms, maintained in continuous culture readily incorporated labelled HCO3- and acetate into amino acids. Labelled propionate, contrast, was utilized only for isoleucine biosynthesis, but failed to label other acids any significant extent. Evidence obtained showing that these mixed, i.e. symbiotic, cultures foward tricarboxylic acid-cycle reactions proceed 2-oxoglutarate. 14C distribution clearly shows 2-oxoglutarate is not oxidized further by enzymes. Instead,...

Complexes of formulation [Cu(Tp(Ph))(L)](ClO(4)) (1-4), where Tp(Ph) is anionic tris(3-phenylpyrazolyl)borate and L N,N-donor heterocyclic base, viz. 2,2'-bipyridine (bpy, 1), 1,10-phenanthroline (phen, 2), dipyridoquinoxaline (dpq, 3), dipyridophenazine (dppz, 4), are prepared from a reaction copper(II) acetate.hydrate with KTp(Ph) in CH(2)Cl(2) isolated as perchlorate salts. The complexes characterized by analytical, structural, spectral methods. crystal structures 1-4 show the presence...
Abstract Retinol acetate hydrolase is localized in rat liver microsomes whereas the retinol palmitate-hydrolyzing activity was found nuclear and mitochondrial lysosome-rich fractions. Acetone-dried powder from whole retained all of its activity. Ammonium sulfate fractionation acetone-dried yielded a partially purified enzyme liberating 240 mµmoles per mg protein 60 min when palmitate used as substrate. The pH optimum 8.6 Km 1.6 x 10-4 m. inhibited by ethanol, acetone, Tween 80 activated bile...

The bgl operon of Escherichia coli is rendered cryptic and uninducible in wild‐type cells by the presence DNA structural elements that negatively regulate transcription. We have carried out a detailed analysis sequences implicated negative regulation. Fine‐structure deletion upstream showed at least two involved silencing promoter. Chemical probing genomic vivo region dyad symmetry, present promoter, hypersensitive to KMnO 4 . detected corresponds potential cruciform structure earlier...
Abstract The back and belly fat of pigs fed a diet containing 20% by wt rapeseed oil (22% erucic acid) for 16 weeks was rendered into oil. This pig fat, which contained 5.6% acid, to male rats in three separate experiments at the weeks. In experiment I compared only Brassica campestris var. Span 4.8% acid. II III, commerical lard 0.2% docosenoic commercial 5.4% free acid added, rapessed There no significant (P<0.01) differences observed level hearts diets oil, or plus However, incidence...
Abstract Rat liver cell suspensions oxidized [1-14C]palmitic acid and [1-14C]palmitoyl-l-carnitine efficiently to CO2. Oxidation of tracer amounts palmitate was inhibited by α-bromo-palmitate (2 x 10-4 m). The oxidation palmitoyl-l-carnitine not affected. α-Bromo-palmitoyl-dl-carnitine did have any effect on the either substrate. inhibition [1-14C]palmitate bromo-acid could be overcome increasing concentration palmitic in incubation medium. mitochondria also but additional palmitate. both...
