ABSTRACT A set of coumarin‐3‐carboxamide analogues were designed, synthesized, and evaluated for their ability to impede pancreatic lipase (PL) activity. Out all the analogues, 5dh 5de demonstrated promising inhibitory activity against PL, as indicated by respective IC 50 values 9.20 11.4 μM, compared Orlistat (IC = 0.97 μM). It was found that analogue inhibited PL in a competitive manner with an inhibition constant ( K i ) 4.504 μM. Additionally, docking analysis validated interactions...

A series of new 2,5-disubstituted arylidene derivatives thiazolidinedione (16a-e, 17a-d, 18a-c) designed using molecular hybridization approach were synthesized, structurally characterized, and explored for their anti-obesity potential via inhibition Pancreatic Lipase (PL). Compound 18a presented the most potent PL inhibitory activity with IC50 = 2.71 ± 0.31 µM, as compared to standard drug, Orlistat (IC50 0.99 µM). Kinetic study revealed reversible competitive mode enzyme by compound an...

Several new coumarin and chromene prototype derivatives have been synthesised evaluated for their ERα ERβ selective activity. Coumarin compounds 18 & 19 were found to be the most active, exhibiting potential antiproliferative activity against both ER +ve −ve breast cancer cell lines. The surprise finding of series, however, are novel III chromenes 45 46, with aroyl substitution at 6th position. Both shown potent lines, promote alkaline phosphatase activity, enhance osteoblast mineralization...

Abstract A series of 1,1′‐biphenyl‐3‐carboxamide and furan‐phenyl‐carboxamide analogs were synthesized using an optimized scheme confirmed by 1 H 13 C nuclear magnetic resonance high‐resolution mass spectrometry techniques. The peptidomimetics screened in vitro to understand the inhibitory potential pancreatic lipase (PL). Analogs assessed for PL activity based on interactions, geometric complementarity, docking score. Among analogs, 9 , 29 24 found have most potent with IC 50 values 3.87,...

A series of novel indole-thiazolidinedione hybrid analogues (7a to 7 u) were synthesised, characterised and evaluated for their potential Pancreatic Lipase (PL) inhibition. Amongst the screened analogues, 7r was found be most active PL inhibitor with an IC50 2.67 µM. Furthermore, enzyme inhibition kinetics study revealed a competitive mode by analogues. This fact confirmed via fluorescence spectroscopy which further suggested presence one binding site synthesized Molecular docking performed...

Abstract Owing to its ability induce cellular senescence, inhibit PCNA, and arrest cell division cycle by negatively regulating CDKs as well being a primary target of p53, p21 is traditionally considered tumor suppressor. Nonetheless, several reports in recent years demonstrated pro-oncogenic activities such apoptosis inhibition cytosolic p21, stimulation motility, promoting assembly cyclin D-CDK4/6 complex. These opposing effects on proliferation, supported the observations inconsistent...
