Aspirin gained tremendous popularity during the 1918 Spanish Influenza virus pandemic, 50 years prior to demonstration of their inhibitory action on prostaglandins. Here, we show that influenza A (IAV) infection, prostaglandin E2 (PGE2) was upregulated, which led inhibition type I interferon (IFN) production and apoptosis in macrophages, thereby causing an increase replication. This role PGE2 not limited innate immunity, because both antigen presentation T cell mediated immunity were also...
Human polymorphonuclear leukocytes (PMNL) convert arachidonic acid (20:4) to a number of dihydroxy metabolites, including leukotriene B4 (LTB,), 5S912Rdihydroxy-6,8,10,14-EEEZ-icosatetraenoic (isomer-1), 5S,12S-dihydroxy-6,8,10,14-EEEZ-icosatetraenoic acid, 5S,l2S-dihydroxy-6,8,10,14-EZEZicosatetraenoic (5SY12S-dh-20:4), 5,6-dihydroxy-7,9,11,14-icosatetraenoic and 5,15-dihydroxy-6,8,11,13-icosatetraenoic acid.LTB4 was synthesized rapidly after stimulation PMNL with the divalent cation...
A novel design for template-selective recognition sites in polymers prepared by molecular imprinting is reported. Molecular imprints were against cAMP that contain a fluorescent dye, trans-4-[p-(N,N-dimethylamino)styryl]-N-vinylbenzylpyridinium chloride, as an integral part of the cavity, thus serving both element and measuring fluorescence detection aqueous media. This molecularly imprinted polymer displays quenching presence cAMP, whereas almost no effect observed structurally similar...
Abstract Human neutrophils and monocytes contain a highly specific dehydrogenase which converts 5-hydroxy-6,8,11,14-eicosatetraenoic acid (5-HETE) to 5-oxo-6,8,11,14-eicosatetraenoic (5-oxo-ETE). We have previously shown that 5-oxo-ETE is potent stimulus of neutrophil calcium levels migration now investigated its effects on human eosinophils. 5-Oxo-ETE eosinophil migration, with significant being detected at concentrations as low 1 nM maximal response microM. The responses elicited by were...
In the present study we investigated mechanism for initial step in formation of these products. We found that 1,500 X g supernatant fraction from human PMNL converts 12-epi-6-truns-LTB4 to its &oxo metabolite which was identified by mass spectrometry and U V spectrophotometry. The latter compound subsequently converted corresponding dihydro-oxo product, further metabolized 6,ll-dihydro-12-epi-6-truns-LTB4, major product after longer incubation times. The 5-hydroxyeicosanoid dehydrogenase...
Abstract Although it is known that septic shock induces immunosuppression, the mechanism for this phenomenon not well understood. Monocytes play a central role in pathophysiology, which also characterized by an increased proportion of natural regulatory T (Treg) cells. We therefore investigated whether Treg could be involved decreased monocyte expression CD14 and HLA-DR observed during shock. demonstrated human inhibit LPS-induced retention CD14. Because loss hallmark apoptosis, suggests...
In patients with cystic fibrosis (CF) and asthma, elevated levels of interleukin-8 (IL-8) are found in the airways. IL-8 is a CXC chemokine that chemoattractant for neutrophils through CXCR1 CXCR2 G protein-coupled receptors. We hypothesized acts directly on airway smooth muscle cells (ASMC) way may contribute to enhanced responsiveness remodeling observed CF asthma. The aim this study was determine whether human ASMC (HASMC) express functional receptors (CXCR1 CXCR2) linked cell contraction...
We recently identified a novel pathway for the metabolism of 5(S)-hydroxy-6,8,11,14-eicosatetraenoic acid (5-HETE) by human neutrophils, resulting in oxidation 5-hydroxyl group to give 5-oxo-6,8,11,14-eicosatetraenoic (5-oxo-ETE) (Powell, W. S., Gravelle, F., and Gravel, S. (1992) J. Biol. Chem. 267, 19233-19241). This is quite specific 5-HETE other eicosanoids containing 5(S)-hydroxyl followed 6-trans double bond. In present study we have shown that 5-oxo-ETE very potent raising cytosolic...
Summary Background The flow cytometric basophil activation test by detection of CD63 expression has been developed as an alternative method for in vitro diagnosis IgE‐mediated reactions to various allergens. Despite promising initial studies, the remains disappointing terms sensitivity. CD203c recently demonstrated a specific marker basophils that is rapidly up‐regulated after allergen challenge sensitized patients. Objective goal present study was compare tests using either or...
Evidence is presented for the existence of a prostaglandin‐F 2α receptor in ovine corpora lutea. Prostaglandin F was specifically bound to particulate fraction this tissue with dissociation constant 0.1μM. Prostaglandins 1α , E 1 and 2 had considerably less affinity whereas prostaglandins A B little or no affinity. From comparison binding constants various related compounds it could be concluded that carboxyl group, three hydroxyl groups 5,6‐ cis double bond (but not 13,14‐ trans bond)...
Abstract 15-Deoxy-Δ12,14-PDJ2 (15d-PGJ2) is a degradation product of PGD2 that has been proposed as an anti-inflammatory compound because its various inhibitory effects, some which are mediated by peroxisome proliferator-activated receptor-γ. In contrast to reported effects on macrophages and other cells, we found this potent activator eosinophils, inducing calcium mobilization, actin polymerization, CD11b expression. It selective for having little or no effect neutrophils monocytes....
Prostaglandin F2alpha was specifically bound by a particulate fraction from bovine corpora lutea. The rate constants for the association (7.5 X 10(3) M-1 S-1) and dissociation (2.1 10-4 reactions gave constant of 2.8 10(-8) M which is similar to that determined Scatchard plot binding data at equilibrium (5 M). receptor stable several hours 23 degrees C but rapidly destroyed 37 C. pH optimum reaction 6.3. had high specificity prostaglandin much lower affinities other prostaglandins....
