Aims: Most chemotherapeutic agents exploit apoptotic signaling to trigger cancer cell death, which frequently results in drug resistance. Necroptosis, a nonapoptotic form of regulated offers an alternative strategy eradicate apoptosis-resistant cells. We previously reported natural necroptosis inducer 2-methoxy-6-acetyl-7-methyljuglone (MAM) A549 lung The current study is designed investigate the detailed necroptotic and its cytotoxicity on drug-resistant Furthermore, vivo anticancer effects...

The incidence of ulcerative colitis (UC), one the two types inflammatory bowel disease, is increasing in many countries. Various natural products have been demonstrated with therapeutic potentials for UC. Herein, effects and mechanisms isobavachalcone (IBC), a chalcone, were evaluated dextran sulfate sodium (DSS)-induced mice lipopolysaccharide (LPS)-stimulated RAW264.7 macrophages. results that IBC treatment significantly improved clinical symptoms, assessed by disease activity index (DAI)...

The anti-cancer effect of high concentrations ascorbic acid (AA) has been well established while its underlying mechanisms remain unclear. association between iron and AA attracted great attention but was still controversial due to the complicated roles in tumors. Our study aims explore interaction cancer. The MTT ATP assays were used evaluate cytotoxicity AA. Reactive oxygen species (ROS) generation, calcium (Ca2+), lipid peroxidation monitored with flow cytometry. Mitochondrial dysfunction...

Nanoscale materials (such as TiO2, hydroxyapatite nanoparticles) have gained much concern in the coating of implants for cell adhesion and growth to improve osteoconductivity. However, due attrition corrosion, wear particles would be generated from joint living organism, influence physiological function synovial membranes cavity. In this study, potential cytotoxicity anatase TiO2 nanoparticles (TiO2 NPs) on rat line 364 (RSC-364) was investigated. After treatment with different...

Blood brain barrier (BBB) disruption is a crucial factor contributing to secondary injury after intracerebral hemorrhage (ICH). Heat shock protein B8 (HSPB8) has been recently reported confer neuroprotection against ischaemic stroke through maintaining BBB integrity. However, the role of HSPB8 in ICH still elusive. In this study, we found that was upregulated by and extensively expressed neurovascular structure including endothelial cells astrocytes. lentivirus intracerebroventricular...

Triptolide, which has been used to treat inflammatory diseases, also reported inhibit proliferation of cancer cells. However, it can cause severe nephrotoxicity, limiting its clinical use. Here, nephrotoxicity triptolide was observed in vivo and vitro. Heat shock protein 72 (HSP72) upregulated during kidney injury rats. HSP72 partially protected human proximal tubule cell lines HK-2 HKC from triptolide-induced injury. Phospho-Raf, phospho-MEK phospho-ERK were elevated cells that...

Psoralidin (PSO) is a natural coumarin isolated from the seeds of Psoralea corylifolia Linn. Previous studies have reported that PSO exerts numerous pharmacological bioactivities including antitumor. The present study aimed to investigate its anticancer effect using colon cancer cells. Cultured HT-29 and HCT-116 cells were treated with different concentrations PSO, cell viability, intracellular reactive oxygen species (ROS), protein expression, apoptosis determined by MTT assay, DCFH2 -DA...

Nine new limonoids, meliazedarines A–I (1–9), seven known analogues (10–16), and five triterpenoids (17–21) were isolated from the fruits of Melia azedarach. Their structures determined by analysis 1D 2D NMR, HRESIMS, X-ray diffraction, electronic circular dichroism (ECD) data. Compound 7 showed significant cytotoxicity against HCT116 cell line with IC50 values 0.3 ± 0.1 μM.

Recently, uncontrolled actin polymerization has been recognized as an initiator of early-onset blood–brain barrier (BBB) rupture. Here, using in vitro models, we found that after oxygen–glucose deprivation/reperfusion (OGD/R), endothelial overexpression HSPB8 suppressed aberrant and thus preserved the integrity BBB. We further investigated mechanisms control assembly. RhoA/ROCK2/p-MLC signaling pathway bEnd.3 cells RhoA activator abrogated inhibitory action on reorganization OGD/R. In...
