Direct chemical modification of native antibodies in a site-specific manner remains great challenge. Ligand-directed conjugation can achieve the selective antibodies, but usually requires multiple extra steps for ligand release and cargo assembly. Herein, we report novel, traceless strategy to enable facile efficient one-step synthesis antibody-drug conjugates (ADCs) by harnessing thioester-based acyl transfer reagent. The designed reagent, consisting an optimized Fc-targeting ligand,...

Although classic Hodgkin lymphoma (cHL) is highly curable with current treatment paradigms, therapy fails in 10-25% of patients. This prospective multicenter phase II study attempted to investigate the efficacy and safety combination tislelizumab gemcitabine oxaliplatin (T-GemOx) relapsed or refractory cHL. Participants received six eight courses (1 g/m2 on day 1) (100 mg/m2 combined (200 mg 2) at 21-day intervals, followed by maintenance (every 2 months for years). The main outcome measure...

Gorham-Stout disease is a life-threatening disorder often manifested by lymphatic malformation and osteolysis. Unfortunately, available therapies are not uniformly effective carry substantial morbidity. We report an 18-year-old male with lytic rib lesions intractable pleural effusion that responded dramatically to the combination of mammalian target rapamycin (mTOR) inhibitor sirolimus aminobisphosphonate zoledronic acid after failing interferon therapy. This tolerable therapeutic has...

Glycosite-specific antibody‒drug conjugatess (gsADCs), harnessing Asn297 N-glycan of IgG Fc as the conjugation site for drug payloads, usually require multi-step glycoengineering with two or more enzymes, which limits substrate diversification and complicates preparation process. Herein, we report a series novel disaccharide-based substrates, reprogram to one-step synthesis gsADCs, catalyzed by an endo-N-acetylglucosaminidase (ENGase) Endo-S2. via ENGases has steps: deglycosylation wild-type...

This paper proposes a new texture classification algorithm that is invariant to rotation and gray-scale transformation. First, we convert two-dimensional (2-D) images one-dimensional (1-D) signals by spiral resampling. Then, use quadrature mirror filter (QMF) bank decompose sampled into subbands. In each band, take high-order autocorrelation functions as features. Features in different bands, which form vector sequence, are then modeled hidden Markov model (BMM). During classification, the...

Objective Ultrasound imaging has emerged as the preferred modality for ovarian tumor screening due to its non-invasive nature and real-time dynamic capabilities. However, in many developing countries, ultrasound diagnosis remains dependent on specialist physicians, where shortage of skilled professionals relatively low accuracy manual diagnoses significantly constrain efficiency. Although deep learning achieved remarkable progress medical image segmentation recent years, existing methods...

The drug-to-antibody ratio (DAR) value and dual-drug combination greatly influence the therapeutic index of antibody-drug conjugates (ADCs). reported approaches usually require multifunctional branched linkers, a complicated technologies, or protein-protein ligation, which may incorporate multihydrophobic fragments result in low coupling efficiency. Herein, we developed facile efficient one-pot method to assemble dual-site-specific ADCs with defined DARs at both N-glycosylation site K248...

Abstract Strain‐promoted azide–alkyne cycloaddition using dibenzoazacyclooctyne (DBCO) is widely applied in copper‐free bioorthogonal reactions. Reported here the efficient acid‐promoted rearrangement and silver‐catalyzed amidation of DBCO, which alters its click reactivity robustly. In switched reaction, as a caged acylation reagent, enables rapid peptide/protein modification after decaging facilitated by silver catalysts, rendering site‐specific conjugation an IgG antibody Fc‐targeting peptide.
