Although much is known about the embryo during implantation, architecture of uterine environment in which early develops not well understood. We employed confocal imaging combination with 3D analysis to identify and quantify dynamic changes luminal structure murine uterus preparation for implantation. When applied mouse mutants implantation defects, this method detected striking peri-implantation abnormalities morphology that cannot be visualized by histology. revealed organization glands...

Intratumoral T cells that might otherwise control tumors are often identified in an "exhausted" state, defined by specific epigenetic modifications and upregulation of genes such as CD38, cytotoxic T-lymphocyte-associated protein 4 (CTLA4), programmed cell death 1 (PD1). Although the term imply inactivity, there has been little study this state at phenotypic level to understand extent their incapacitation. Starting with observation move more quickly through mouse longer they reside progress...

Abstract Multiphoton microscopy is a powerful technique for deep in vivo imaging scattering samples. However, it requires precise, sample-dependent increases excitation power with depth order to generate contrast tissue, while minimizing photobleaching and phototoxicity. We show here how adaptive can optimize illumination at each point 3D volume as function of the sample’s shape, without need specialized fluorescent labeling. Our method relies on training physics-based machine learning model...

The nucleolus, a membraneless organelle crucial for ribosome production, has unique nanoscale structure whose organization is responsive to cell signals and disease progression. Here, we highlight the potential of Expansion Microscopy (ExM) capturing intricate spatial functional information about organelles such as nucleolus nuclear foci. We apply dual protein (dual-proExM) in combination with click (click-ExM) capture images at highest resolution reported ∼45 ± 2 nm. Inhibition nucleolar...

Abstract Regulatory T cells (Tregs) reside in nonlymphoid tissues where they carry out unique functions. The molecular mechanisms responsible for Treg accumulation and maintenance these are relatively unknown. Using an unbiased discovery approach, we identified LAYN (layilin), a C-type lectin-like receptor, to be preferentially highly expressed on subset of activated Tregs healthy diseased human skin. Expression layilin was induced by TCR-mediated activation the presence IL-2 or TGF-β. Mice...
