Cultured human umbilical vein endothelial cells (HUVEC) stimulated with thrombin are known to synthesize prostacyclin at least in part from arachidonate released by phospholipase A2, an enzyme directly activated calcium. In this study, stimulation of Quin 2-loaded HUVEC caused rapid and dose-dependent rises inositol trisphosphate (IP3) cytosolic free calcium (Ca2+i) levels which preceded a similarly rise production measured as 6-ketoprostaglandin F1 alpha (6-keto-PGF1 alpha) radioimmunoassay...

The α-helix structures of the anti-HIV fusion inhibitory peptides are stabilized by amino acid sequence and intrachain hydrogen bonds. study peptide analogues using (E)-alkene (Z)-fluoroalkene dipeptide isosteres demonstrated substantial, yet position-dependent, contribution bonds to stability bioactivity.

ABSTRACT Peptides derived from the α-helical domains of human immunodeficiency virus (HIV) type 1 (HIV-1) gp41 inhibit HIV-1 fusion to cell membrane. Enfuvirtide (T-20) is a peptide-based drug that targets step HIV fusion, and as such, it effectively suppresses replication strains are either wild or resistant multiple reverse transcriptase and/or protease inhibitors. However, variants with T-20 resistance have emerged; therefore, development new potent inhibitors urgently needed. We...

Abstract Although hepatoblastoma is the most common pediatric liver cancer, its genetic heterogeneity and therapeutic targets are not well elucidated. Therefore, we conducted a multiomics analysis, including mutatome, DNA methylome, transcriptome analyses, of 59 samples. Based on methylation patterns, was classified into three clusters exhibiting remarkable correlation with clinical, histological, features. Cluster F largely composed cases fetal histology good outcomes, whereas E1 E2...

KMT2A-rearranged infant acute lymphoblastic leukemia (ALL) represents the most refractory type of childhood leukemia. To uncover molecular heterogeneity this disease, we perform RNA sequencing, methylation array analysis, whole exome and targeted deep sequencing on 84 infants with Our multi-omics clustering followed by single-sample single-cell inference hematopoietic differentiation establishes five robust integrative clusters (ICs) different master transcription factors, fusion partners...

Abstract Novel therapeutic strategies are urgently required for osteosarcoma, given the early age at onset and persistently high mortality rate. Modern transcriptomics techniques can identify differentially expressed genes (DEGs) that may serve as biomarkers targets, so we screened DEGs in osteosarcoma. We found osteosarcoma cases could be divided into fair poor survival groups based on gene expression profiles. Among upregulated group, siRNA-mediated knockdown of glycosylation-related...

Reported herein are the design, biological activities, and biophysical properties of a novel HIV-1 membrane fusion inhibitor. α-Helix-inducible X-EE-XX-KK motifs were applied to design an enfuvirtide analogue 2 that exhibited highly potent anti-HIV activity against wild-type HIV-1, enfuvirtide-resistant strains, HIV-2 strain in vitro. Indispensable residues for bioactivity enfuvirtide, including interacting with N-terminal heptad repeat C-terminal hydrophobic residues, identified.

Enfuvirtide (T-20) is a fusion inhibitor that suppresses replication of human immunodeficiency virus (HIV) variants with multi-drug resistance to reverse transcriptase and protease inhibitors. It peptide derived from the C-terminal heptad repeat (C-HR) HIV-1 gp41, it prevents interactions between C-HR N-terminal HR (N-HR) thus interfering conformational changes are required for viral fusion. However, prolonged therapies T-20 result in emergence T-20-resistant strains contain primary...
