The only molecular similarity in sex determination found so far among phyla is between the Drosophila doublesex (dsx) and Caenorhabditis elegans mab-3 genes. dsx contain a zinc finger-like DNA-binding motif called DM domain, perform several related regulatory functions, are at least partially interchangeable vivo. A domain gene Dmrt1 has been implicated male gonad development variety of vertebrates, on basis embryonic expression chromosomal location. Such evidence highly suggestive conserved...
DNA site-specific recombinases (SSRs) such as Cre, FLPe, and φC31, are powerful tools for analyzing gene function in vertebrates. While the availability of multiple high-efficiency SSRs would facilitate a wide array genomic engineering possibilities, efficient recombination mammalian cells has only been observed with Cre recombinase. Here we report de novo synthesis mouse codon-optimized FLP (FLPo) ΦC31 (ΦC31o) SSRs, which result efficiencies similar to Cre.
Deletion of the distal short arm chromosome 9 (9p) has been reported in a number cases to be associated with gonadal dysgenesis and XY sex reversal, suggesting that this region contains one or more genes required two copies for normal testis development. Recent studies have greatly narrowed interval containing putative autosomal testis-determining gene(s) portion 9p24.3. We previously identified DMRT1, human gene sequence similarity regulate sexual development nematodes insects. These...
Abstract Gene trap mutagenesis in embryonic stem cells is an important tool to help elucidate gene function current mouse efforts. Vector systems based on inversion of the module have recently been devised allow for conditional mutagenesis. However, additional efforts are needed improve this technology including improving efficiency site‐specific recombinases required manipulate these vectors vivo. Here we describe a line carrying codon‐optimized FLP recombinase Flpo at ROSA26 locus that...
Summary: Vertebrates employ varied strategies, both chromosomal and nonchromosomal, to determine the sex of developing embryo. Among reptiles, temperature-dependent determination (TSD) is common. The temperature incubation during a critical period preceding sexual differentiation determines future embryo, presumably by altering activity or expression regulatory factor(s). Here we examine Dmrt1 gene, candidate regulator mammalian avian development, in turtle. During sex-determining period,...
This study evaluates a computer clustering procedure for predicting the outcome of psychiatric patients. Premorbid and performance evaluations patients were obtained during hospitalization. Active treatment led to relatively high discharge rate. The procedures successful predictions adjustment levels which differed significantly three schizophrenic groups. Performance clusters correlated higher than premorbid with adjustment. Implications data-based mental health planning evaluation are discussed.
Teratogenic potential of human drugs is assessed in embryo-fetal development (EFD) studies two species as per regulatory guidelines. In vitro developmental toxicity assays can be vital early drug efforts to distinguish teratogenic drugs, while reducing animal use. Results from (rat whole embryo culture assay and mouse embryonic stem cell test), were evaluated for their ability predict the 83 compounds with known EFD outcome rats. With an integrated model, sensitivity specificity teratogens...
A method for purifying IgM monoclonal antibodies (mAbs) from murine ascites using a DEAE-disk is described. After ammonium sulfate precipitation, proteins are redissolved and loaded onto DEAE-disk. MAb then eluted the disk stepwise NaCl gradient. mAb produced by this procedure was >95% pure as assessed reducing SDS-PAGE analysis free of significant IgG contamination determined double radial immunodiffusion analysis. Yield ̃2% total protein ̃10% amount contained in crude ascites. retained...
An enhanced understanding of the microstructure oxide ceramics will help scientists and engineers improve their efficiency design. A phase-field model for composition phase distribution ceramic components is studied. The model, which includes an obstacle in portion energy potential, results a minimisation problem that characterises bulk phases. transition region between them studied several mathematically plausible asymptotic limits. behaviour system these limits provides insights into...
The standard of care treatment for neovascular age-related macular degeneration, delivered as ocular injection, is based on anti-vascular endothelial growth factor (anti-VEGF). course may need to be modified quickly certain patients their response. Models that track both the concentration and response an anti-VEGF are presented. specific focus assess existence analytical solutions different types models. Both ODE-based model a map-based illustrate dependence solution various biological...
