The technique of recording His bundle electrograms in man by a tripolar electrode catheter was applied study patients with first, second, and third-degree heart block. In all first-degree block (congenital, acquired, or induced atrial pacing), the delay A-V conduction occurred proximal to as evidenced prolonged P-H interval. enhancement (shortening interval) due isoproterenol qualitatively similar three forms cases Wenckebach phenomenon interval progressively increased until dropped beat...

Abstract Cell-free DNA (cfDNA) has the potential to inform tumor subtype classification and help guide clinical precision oncology. Here we develop Griffin, a framework for profiling nucleosome protection accessibility from cfDNA study phenotype of tumors using as low 0.1x coverage whole genome sequencing data. Griffin employs GC correction procedure tailored variable fragment sizes, which generates better representation chromatin improves accuracy cancer detection classification. We...

Preeclampsia is characterized by placental dysfunction and results in significant morbidity, but reliable early prediction remains challenging. We investigated whether clinically obtained prenatal cell-free DNA (cfDNA) screening (PDNAS) using whole-genome sequencing (WGS) data can be leveraged to predict preeclampsia risk pregnancy (≤16 weeks). Using 1,854 routinely collected clinical PDNAS samples (median, 12.1 weeks) with low-coverage (0.5×) WGS data, we developed a framework quantify...

The exon junction complex (EJC) deposited upstream of mRNA junctions shapes structure, composition, and fate spliced ribonucleoprotein particles (mRNPs). To achieve this, the EJC core nucleates assembly a dynamic shell peripheral proteins that function in diverse post-transcriptional processes. illuminate consequences composition change, we purified EJCs from human cells via RNPS1 CASC3. We show originates as an SR-rich mega-dalton-sized RNP contains but lacks Sometime before or during...

Advanced prostate cancers comprise distinct phenotypes, but tumor classification remains clinically challenging. Here, we harnessed circulating DNA (ctDNA) to study phenotypes by ascertaining nucleosome positioning patterns associated with transcription regulation. We sequenced plasma ctDNA whole genomes from patient-derived xenografts representing a spectrum of androgen receptor active (ARPC) and neuroendocrine (NEPC) cancers. Nucleosome transcriptional activity were reflected in at regions...

The least common type response observed that in which A-V delay limited only to nodal region (type A). This obtained whenever increased direct proportion prematurity a propagated impulse. It characterized by progressive prolongation P-H interval with ultimate failure occurring...

In eukaryotic cells, proteins that associate with RNA regulate its activity to control cellular function. To fully illuminate the basis of function, it is essential identify such RNA-associated proteins, their mode action on RNA, and preferred targets binding sites. By analyzing catalogs human defined by ultraviolet light (UV)-dependent -independent approaches, we classify these into two major groups: (i) widely recognized (RBPs), which bind directly UV-crosslink efficiently (ii) a new group...

Thus, zone concealment could be localized region proximal bundle. The clinical findings confirmed in three animal experiments. nonconducted P waves also blocked aberrant beats distinguished from presence preceding...

Abstract The Exon Junction Complex (EJC) deposited upstream of exon-exon junctions during pre-mRNA splicing in the nucleus remains stably bound to RNA modulate mRNA fate at multiple post-transcriptional steps until its disassembly translation. Here, we investigated two EJC mechanisms human embryonic kidney 293 (HEK293) cells, one mediated by PYM1, a factor that can bind both ribosome and RBM8A/MAGOH heterodimer core, another elongating itself. We find EJCs lacking PYM1 interaction show no...

Many post-transcriptional mechanisms operate via mRNA 3′UTRs to regulate protein expression, and such controls are crucial for development. We show that homozygous mutations in two zebrafish exon junction complex (EJC) core genes rbm8a magoh leads muscle disorganization, neural cell death, motor neuron outgrowth defects, as well dysregulation of mRNAs subjected nonsense-mediated decay (NMD) due translation termination ≥ 50 nts upstream the last exon-exon junction.
