Abstract Alzheimer's disease is a complex progressive neurodegenerative disorder with profound cognitive decline. Multiple susceptibility genetic variants have been identified equivocal replication. While rare, collections of extended pedigrees multiple affected family members are invaluable for genome‐wide screens. We used two pedigrees, having 14–15 siblings four to five late‐onset patients in each, identify the gene, transient receptor potential cation channel, subfamily C, member 4...

Background: Alzheimer's disease is complex, with variants in multiple genes contributing to interactions increasing risk for the disease. Brain-derived neurotrophic factor (BDNF) promotes neuronal survival and modulates hippocampal-dependent memory. Methods: We examined 11 SNPs that spanned gene on chromosome 11p14 220 patients 128 control spouses. Results: Not all of were informative, due minor allele frequencies <2%. Neither C270T nor two reside proximal exon V had...

Neuregulin 4 (Nrg4), a newly identified adipokine secreted by brown adipose tissue, is hypothesised to play crucial role in metabolism. The present study aimed evaluate the association between serum Nrg4 levels and non-alcoholic fatty liver disease (NAFLD) children with obesity China. A total of 123 were included this study. Anthropometric biochemical parameters measured all subjects. NAFLD was diagnosed using ultrasonography. Nrg4, leptin adiponectin enzyme-linked immunosorbent assay. 58...

Gestational diabetes mellitus (GDM) “program” an elevated risk of metabolic syndrome in the offspring. Epigenetic alterations are a suspected mechanism. GDM has been associated with placental DNA methylation changes some epigenome-wide association studies. It remains unclear which genes or pathways affected, and whether any differential gene methylations correlated to fetal growth circulating health biomarkers. In study using Infinium MethylationEPIC Beadchip, we sought identify genome-wide...

It remains unclear which early gestational biomarkers can be used in predicting later development of diabetes mellitus (GDM). We sought to identify the optimal combination GDM machine learning (ML) models.

Dexmedetomidine is a highly specific alpha2-adrenergic agonist, which used clinically as an anesthetic adjuvant and in animal experiments has neuroprotective effect during ischemia. The current study showed that dexmedetomidine enhances glutamine disposal by oxidative metabolism astrocytes. This occurs at pharmacologically relevant concentrations. It exerted on receptors not imidazoline-preferring sites, it large enough to reduce the availability of precursor neurotoxic glutamate.

Cystatin C, a protease inhibitor with widespread distribution, is upregulated in response to injury. Levels are elevated the brains of patients Alzheimer disease (AD). We compared frequencies for CST 3 exon 1 polymorphism AD and controls. A proportional odds model indicated that APOE4 combination carried high risk: 14-fold elevation men 16-fold women. These risks apply risk at ages older than 64 years shift onset younger 65 years.

Abstract Background Fetal overgrowth “programs” an elevated risk of type 2 diabetes in adulthood. Epigenetic alterations may be a mechanism programming the vulnerability. We sought to characterize genome-wide placental gene methylations fetal and associations with metabolic health biomarkers including leptin, adiponectin growth factors. Results Comparing DNA large-for-gestational-age (LGA, indicator overgrowth, n = 30) versus optimal-for-gestational-age (OGA, control, infants using Illumina...
