In order to characterize the thermodynamic constraints on process of integral membrane protein folding and assembly, we have conducted a biophysical dissection structure bacteriorhodopsin (BR), prototypical α-helical protein. Seven polypeptides were synthesized, corresponding each seven transmembrane α-helices in BR, individual polypeptide was characterized reconstituted phospholipid vesicles. Five form stable isolation from remainder tertiary BR. However, using our reconstitution protocols,...

The two major anisocratic elution modes were compared in reversed-phase chromagraphy with 180 μm I.D. fused-silica capillary columns acked 6 Zorbax SB ODS-silica. By evaluating the retention factors of alkylbenzenes at acetonitrile concentrations varying from 60 to 80% (v/v) aqueous eluent and temperature range 30–80°C, it was found that a 5°C change column 1% concentration have almost same effect on retention. This is illustrated by identical chromatograms an alkylbenzene sample obtained...

Abstract Catecholamines and their metabolites have been separated isocratically by reverse-phase chromatography with aqueous (no organic solvent admixed) eluents. Unlike ion-exchange or ion-pair chromatography, mixtures of both acidic basic substances can be in a single chromatographic run, because the retention is governed hydrophobic interactions between nonpolar moiety solute molecules octadecyl-silica stationary phase. The relative values strongly depend on pH eluent, which governs...
