See related commentary by Kanis, page [1829][1] Since the publication of Osteoporosis Canada guidelines in 2002, there has been a paradigm shift prevention and treatment osteoporosis fractures. [1][2],[2][3] The focus now is on preventing fragility fractures their negative
The organization of cerebral networks was estimated within individuals with intensive, repeat sampling fMRI data. A hierarchical emerged in each individual that delineated first-, second-, and third-order cortical networks. Regions distinct association consistently exhibited side-by-side juxtapositions repeated across multiple zones, clear robust functional specialization among the embedded regions.
Proton pump inhibitor (PPI) use has been identified as a risk factor for hip and vertebral fractures. Evidence supporting relationship between PPI osteoporosis remains scant. Demonstrating that PPIs are associated with accelerated bone mineral density (BMD) loss would provide supportive evidence mechanism through which could increase fracture risk.We used the Canadian Multicentre Osteoporosis Study data set, enrolled population-based sample of Canadians who underwent BMD testing femoral...
We aimed to explore whether frailty was associated with fracture risk and could modify the propensity of type 2 diabetes toward increased fractures.Data were from a prospective cohort study. Our primary outcome time first incident clinical fragility fracture; secondary outcomes included hip spine fracture. Frailty status measured by Index (FI) deficit accumulation. The Cox model incorporating an interaction term (frailty × diabetes) used for analyses. The analysis 3,149 (70% women)...
Mortality rates from chronic kidney disease (CKD) have increased in the last decade. In this pre-specified analysis of DAPA-CKD trial, we determined effects dapagliflozin on cardiovascular and non-cardiovascular causes death.DAPA-CKD was an international, randomized, placebo-controlled trial with a median 2.4 years follow-up. Eligible participants were adult patients CKD, defined as urinary albumin-to-creatinine ratio (UACR) 200-5000 mg/g estimated glomerular filtration rate (eGFR) 25-75...
ABSTRACT We compared two methods for osteoporotic vertebral fracture (VF) assessment on lateral spine radiographs, the Genant semiquantitative (GSQ) technique and a modified algorithm-based qualitative (mABQ) approach. evaluated 4465 women 1771 men aged ≥50 years from Canadian Multicentre Osteoporosis Study with available X-ray images at baseline. Observer agreement was lowest grade 1 VFs determined by GSQ. Among physician readers, greater diagnosed mABQ (ranging 0.62 [95% confidence...
This pre-specified analysis of DAPA-CKD assessed the impact sodium-glucose cotransporter 2 inhibition on abrupt declines in kidney function high-risk patients based having chronic disease (CKD) and substantial albuminuria. was a randomized, double-blind, placebo-controlled trial that had median follow-up 2.4 years. Adults with CKD (urinary albumin-to-creatinine ratio 200–5000 mg/g estimated glomerular filtration rate 25–75 mL/min/1.73m2) were randomized to dapagliflozin 10 mg/day matched...
The warm-up programme “FIFA 11+” has been shown to reduce football injuries in different populations, but so far veteran players have not investigated. Due differences age, skill level and gender, a simple transfer of these results is recommended. purpose this study was investigate the preventive effects players.Twenty teams were recruited for prospective 9-month (1 season) cluster-randomised trial. intervention group (INT, n = 146; 45 ± 8 years) performed at beginning each training session,...
Data on long-term consequences of non-hip non-vertebral (NHNV) fractures, accounting for approximately two-thirds all fragility are scanty. Our study aimed to quantify the population-wide impact NHNV fractures mortality. The national population-based prospective cohort (Canadian Multicentre Osteoporosis Study) included 5526 community dwelling women and 2163 men aged 50 years or older followed from July 1995 September 2013. Population number was used average people whom one death would be...
Osteoporosis Canada's 2010 Clinical Practice Guidelines for the Diagnosis and Management of in Canada focus on clinical impact fragility fractures, assessment management women men at high risk fracture. These guidelines now integrate a 10-year absolute fracture prediction into an overall approach by using validated tools. There currently is large gap between optimal practices those that are being provided to Canadians with osteoporosis. part concerted effort close this gap. Key changes from...
The human cerebral cortex is populated by specialized regions that are organized into networks. Here we estimated networks using a Multi-Session Hierarchical Bayesian sModel (MS-HBM) applied to intensively sampled within-individual functional MRI (fMRI) data. network estimation procedure was initially developed and tested in two participants (each scanned 31 times) then prospectively 15 new 8 11 times). Detailed analysis of the revealed global organization. Locally first-order sensory motor...
Diet-derived lipids may influence cytokine-mediated endothelial cell dysfunction, including TNF-induced apoptosis. To test this hypothesis, oxidative stress, intracellular calcium levels, barrier function, viability, and apoptosis were measured in vascular cells treated with 90 microM linoleic acid (18:2, n-6) and/or 20 ng/mL TNF (100 U/mL). For short-term exposure, exposed to 18:2 for 6 h or 1.5 h. long-term cultures 24 19.5 In + TNF, pretreatment began 4.5 before additional exposure either...
Haemolysin Kanagawa, a toxin from Vibrio parahaemolyticus, is known to trigger haemolysis. Flux studies indicated that haemolysin forms cation channel. In the present study, channel properties were elucidated by patch clamp and functional significance of ion fluxes fluorescence-activated cell sorting (FACS) analysis. Treatment human erythrocytes with 1 U ml−1 within minutes induces non-selective permeability. Moreover, activates clotrimazole-sensitive K+ channels, pointing stimulation...
