We report the discovery and medicinal chemistry optimization of a novel series pyrazole-based inhibitors human lactate dehydrogenase (LDH). Utilization quantitative high-throughput screening paradigm facilitated hit identification, while structure-based design multiparameter enabled development compounds with potent enzymatic cell-based inhibition LDH activity. Lead such as 63 exhibit low nM both LDHA LDHB, submicromolar production, glycolysis in MiaPaCa2 pancreatic cancer A673 sarcoma...
A synthetic approach to paxilline indole diterpenes is described. The route the pentacyclic core relies on a new regioselective alkenylation of ketones and tandem radical addition–aldol reaction sequence access vicinal quaternary stereocenters. Emindole SB, simplest member family, synthesized in 11 steps from commercially available material demonstrate application this approach.
Ascospores of Neosartorya, Byssochlamys and Talaromyces can be regarded as the most stress-resistant eukaryotic cells. They survive exposure at temperatures high 85°C for 100 min or more. Neosartorya fischeri ascospores are more viscous resistant to combined stress heat desiccation than macrosporus which contain predominantly trehalose. These trehalose-based oligosaccharides (TOS) that novel compatible solutes, accumulated levels. compounds also found in other members genus some genera...
Paxilline indole diterpenes have been a subject of scientific inquiry for over 30 years due to their diverse biological activities and unique structural features. These efforts resulted in multitude synthetic approaches dozen completed synthesis. Here, we discuss our contributions this area that culminated an 11-step synthesis (±)-emindole SB.
Three complex α-1,6-linked trehalose-based oligosaccharides with unique preservation properties, isobemisiose, neosartose, and fischerose, were recently identified from the extreme stress-tolerant ascospores of Neosartorya fischeri. Herein, we report first concise, scalable, iterative chemical synthesis these a differentially protected thioglycoside donor selectively protected, asymmetric trehalose acceptor. This work constitutes an improved is also reported tetrasaccharide, pentasaccharide,...
