Long Covid is a debilitating condition of unknown etiology. In pDCs, BTK-inhibition (BTKi) blocks the IFN-α production via TLR-9, but not TLR-7. Upon TLR-7 stimulation, BTKi enhances of by blocking inhibitory BDCA-2 pathway. This might explain partially failure in SLE and is interest for trials multiple sclerosis.
