Effects of Honghua preserved amniotic membrane on scar healing in experimental glaucoma surgery.

ABSTRACT
To investigate the efficacy and safety of Honghua preserved amniotic membrane (AM) for preventing scar formation filtering bleb in a rabbit model glaucoma trabeculectomy surgery.Totally 36 rabbits (36 eyes) were randomly divided into 3 groups: experimental group (ocular combination with AM transplantation), control surgery implantation), blank (single trabeculectomy). Clinical observations [including intraocular pressure (IOP), blebs complications], Masson-Trichrome staining, real-time quantitative reverse transcription-polymerase chain reaction (real-time PCR), Western blot performed on different time points (D1, D7, D14, D21 D56) after surgery.After operated 14d, there statistically significant differences compared to situation before operation (P<0.05), whereas no difference that among three groups (P>0.05). After 21d, IOP was lowest (P<0.05). There between On postoperative mean number fibroblasts significantly lower (40.6±10.2) those (54.4±10.8) (68.2±11.6) (P<0.05, respectively). The numbers macrophage respcitively versus P<0.05, Compared group, level transforming growth factor-β (TGF-β1) expression sclera conjunctival areas reduced protein mRNA but not these two (P>0.05).The trabeculectory can IOP, sustain functional filtration bleb, inhibit proliferation open filtrating pathway models.
