The World Health Organization (WHO) has declared the coronavirus disease 2019 (COVID-19) as an international health emergency. Current diagnostic tests are based on reverse transcription-quantitative polymerase chain reaction (RT-qPCR) method, which is gold standard test that involves amplification of viral RNA. However, RT-qPCR assay limitations in terms sensitivity and quantification. In this study, we tested both qPCR droplet digital PCR (ddPCR) to detect low amounts cycle threshold (CT)...

Abstract Rift valley fever (RVF) is an important zoonotic disease caused by the virus (RVFV) which can affect ruminants and humans. In this study, a comparison was done of reverse transcription-quantitative polymerase chain reaction (RT-qPCR) transcription-droplet digital PCR (RT-ddPCR) assays with synthesized RVFV RNA, cultured viral mock clinical RNA samples. The genomic segments (L, M, S) three strains (BIME01, Kenya56, ZH548) were used as templates for in vitro transcription (IVT). Both...

Rapid and accurate sequencing covering the entire genome is essential to identify genetic variations of viral pathogens. However, due low titers in clinical samples, certain amplification steps are required for sequencing. At present, there no universal primers available alphacoronaviruses that, since these viruses have diverse strains, new specific target strain must be continuously developed Thus, this study, we aimed develop a primer set valid all human applicable samples containing trace...

As SARS-CoV-2 variants of concern emerged, the genome sequencing strains became more important. In this study, was sequenced using amplicon-based with MinION. The primer panel used in study consisted only 11 panels and size amplicons approximately 3 kb. Full sequences were obtained a hundred copies genome, 92.33% 75.39% 10 genome. few differences nucleotide originated from mutations laboratory cultures and/or mixed sequences. quantification genomic RNA done RT-ddPCR methods, level LoD...

The human rhinovirus (RV) is a positive-stranded RNA virus that causes respiratory tract diseases affecting both the upper and lower halves of system. RV enhances its replication by concentrating synthesis within modified host membrane in an intracellular compartment. infections often occur alongside caused other viruses, may remain asymptomatic for extended periods. Alongside qualitative detection, it essential to accurately quantify from clinical samples explore relationships between viral...

Snow covers a large surface area of the Earth and provides for exchange biological chemical components. However, microbial composition components snow are poorly understood. We assessed bacterial fungal diversity characteristics in freshly deposited snowfall samples collected from sub-urban site Daejeon, Republic Korea. analyzed using DNA amplification followed by Illumina MiSeq Sequencing microbiome, ion chromatography cations (Na+, Ca2+, Mg2+, NH4+) anions (SO42−, NO3−, Cl−), water-soluble...
