Significance The development of selective bioactive compounds that target RNA structures is difficult. Here we report an approach to identify low molecular weight fragments bind a cancer-causing RNA. By determining where the within target, they can be assembled provide high-affinity and potent inhibitor. This could general way develop small molecules RNA, which previously was considered “undruggable.” It applied precision lead medicine against contributes cancer metastasis.

Natural disasters have frequently occurred and caused great harm. Although the remote sensing technology can effectively provide disaster data, it still needs to consider relevant information from multiple aspects for analysis. It is hard build an analysis model that integrate large-scale information, particularly at sematic level. The antimalarial drug artemisinin is found to have diverse biological activities ranging from anti-inflammatory anticancer properties; however, as of today, the cellular targets and mechanism action this important compound remained elusive. Here, we report global protein target profiling in HeLa cancer cell proteome using a chemical proteomics approach. In presence hemin, multiple proteins were targeted by probe through covalent modification. Further studies revealed that reducing hemin heme...

Significance Because of the weak interactions between individual glycan-binding proteins (GBPs), such as galectin-3, and glycans, strategies that enable direct interrogation these in living cells remain limited. Thus, glycan glycoprotein ligands are physiologically relevant for galectin-3 binding insufficiently described. Here we used a proximity labeling approach catalytically tags interactors identified its pertinent counter-receptors live cells. This study demonstrates is powerful tool...

Forest fires have frequently occurred and caused great harm to people’s lives. Many researchers use machine learning techniques predict forest by considering spatio-temporal data features. However, it is difficult efficiently obtain the features from large-scale, multi-source, heterogeneous data. There a lack of method that can effectively extract required learning-based fire predictions multi-source This paper proposes prediction integrates knowledge graphs models. fuse constructing...

Colorectal cancer remains the second leading cause of cancer-related mortalities worldwide. While artemisinin (ART), a key active compound from traditional Chinese medicinal herb Artemisia annua, has been recognized for its antiproliferative activity against colon cells, underlying molecular underpinnings remain elusive. Whereas promiscuity heme-dependent alkylating macromolecules, mainly proteins, seen pivotal as universal and primary mode action ART in accumulating evidence suggests...

Abstract Rifamycin-derived drugs, including rifampin, rifabutin, rifapentine, and rifaximin, have long been used as first-line therapies for the treatment of tuberculosis other deadly infections. However, late steps leading to biosynthesis industrially important rifamycin SV B remain largely unknown. Here, we characterize a network reactions underlying SV, S, L, O, B. The two-subunit transketolase Rif15 cytochrome P450 enzyme Rif16 are found mediate, respectively, unique C–O bond formation...

Abstract Folates are indispensable for plant development, but their molecular mode of action remains elusive. We synthesized a probe, “5-F-THF-Dayne,” comprising 5-formyl-tetrahydrofolate (THF) coupled to photoaffinity tag. Exploiting this probe in an affinity proteomics study Arabidopsis thaliana, we retrieved 51 hits. Thirty interactions were independently validated with vitro expressed proteins bind 5-F-THF high or low affinity. Interestingly, the interactors reveal associations beyond...
