Edible sea cucumbers are widely used as a health food and medicine. A fucosylated glycosaminoglycan (FG) was purified from the high-value cucumber Stichopus herrmanni. Its physicochemical properties structure were analyzed characterized by chemical instrumental methods. Chemical analysis indicated that this FG with molecular weight of ∼64 kDa is composed N-acetyl-d-galactosamine, d-glucuronic acid (GlcA), l-fucose. Structural clarified contains chondroitin sulfate E-like backbone, mostly...

Sulfated polysaccharides from sea cucumbers possess distinct chemical structure and various biological activities. Herein, three types of were isolated purified Pattalus mollis, their structures bioactivities analyzed. The fucosylated glycosaminoglycan (PmFG) had a CS-like backbone composed the repeating units {-4-d-GlcA-β-1,3-d-GalNAc4S6S-β-1-}, branches sulfated α-l-Fuc (including Fuc2S4S, Fuc3S4S Fuc4S with molar ratio 2:2.5:1) linked to O-3 each d-GlcA. fucan sulfate (PmFS) consisting...

Abstract Fucosylated chondroitin sulfate (FuCS) is a structurally distinct glycosaminoglycan, and its oligosaccharides exhibit excellent anticoagulant activity with lower risks of adverse effects bleeding. Herein we report facile approach to the synthesis FuCS hexa‐ nonasaccharides on basis enzymatic degradation over 12 linear steps. As compared clinical low‐molecular‐weight heparin drug (enoxaparin), nonasaccharide synthesized in this study displayed similar APTT selective intrinsic factor...

Abstract A native fucosylated glycosaminoglycan from sea cucumber Holothuria fuscopunctata (nHG), mainly branched with Fuc3S4S, exhibited potent anticoagulant activity by intrinsic tenase iXase (FIXa-FVIIIa complex) and antithrombin-dependent factor IIa (FIIa) inhibition, but also had the effects of FXII activation platelet aggregation. For screening a selective inhibitor, depolymerized nHG (dHG-1 ∼ –6) pure octasaccharide (oHG-8) were prepared. Like nHG, dHG-1 –6 oHG-8 could potently...

Fucoidan, a sulfated glycan derived from brown algae, has garnered significant attention for its anticoagulant properties. However, the structural complexity and heterogeneity of naturally extracted fucoidan have hindered comprehensive understanding structure-activity relationship, limiting development fucoidan-based drugs. To address this challenge, we synthesize diverse library 58 distinct fucoidans with multiple contiguous 1,2-cis glycosidic bonds, ranging disaccharides to...

Glycosaminoglycans (GAGs) are conserved polysaccharides composed of linear repeating disaccharides and play crucial roles in multiple biological processes animal kingdom. However, saccharide-branched GAGs rarely found, except the fucose-branched one from sea cucumbers. There was conjecture about presence disaccharide-branched GAG since 30 years ago, though not yet confirmed. Here, we report a containing galactose-fucose branches Thelenota ananas. This unique branch confirmed as...
