In order to search for sulfated polysaccharides in different invertebrate connective tissues and examine their biological activities, we have isolated three types of from the body wall sea cucumbers Holothuria edulis, Apostichopus japonicas nobilis. The physicochemical properties anticoagulant activities these were examined compared. chemical composition analysis nuclear magnetic resonance (NMR) indicate that two polysaccharides, fucan fucosylated chondroitin sulfate (FuCS), found all...

Significance Selective inhibition of the intrinsic coagulation pathway is a promising strategy for developing safer anticoagulants without serious bleeding consequences. We prepared and identified series oligosaccharides as inhibitors tenase, which final rate-limiting enzyme complex in an attractive but less explored target due to lack pure selective inhibitor. Analysis these purified reveals precise structure fucosylated glycosaminoglycan. Among oligosaccharides, nonasaccharide minimum...

Polycystin-2 functions as a cation-permeable transient receptor potential ion channel in kidney epithelial cells and when mutated results human autosomal dominant polycystic disease. For further exploration of the in vivo Polycystin-2, this study examined its expression function during zebrafish embryogenesis. pkd2 mRNA is ubiquitously expressed, presence larval could be confirmed by reverse transcription–PCR on isolated pronephroi. Immunostaining with anti-zebrafish antibody...

Sulfated fucans, the complex polysaccharides, exhibit various biological activities. Herein, we purified two fucans from sea cucumbers Holothuria edulis and Ludwigothurea grisea. Their structures were verified by means of HPGPC, FT-IR, GC–MS NMR. As a result, novel structural motif for this type polymers is reported. The have unique structure composed central core regular (1→2) (1→3)-linked tetrasaccharide repeating units. Approximately 50% units L. grisea (100% H. fucan) contain sides...

Edible sea cucumbers are widely used as a health food and medicine. A fucosylated glycosaminoglycan (FG) was purified from the high-value cucumber Stichopus herrmanni. Its physicochemical properties structure were analyzed characterized by chemical instrumental methods. Chemical analysis indicated that this FG with molecular weight of ∼64 kDa is composed N-acetyl-d-galactosamine, d-glucuronic acid (GlcA), l-fucose. Structural clarified contains chondroitin sulfate E-like backbone, mostly...

Enzyme-assisted extraction optimization, characterization and in vitro antioxidant activity of polysaccharides from sea cucumber Phyllophorus proteus (PPP) were investigated the present study. The optimal conditions with a yield 6.44 ± 0.06% for PPP determined as follows: Extraction time 2.89 h, ratio solvent to raw material 16.26 mL/g, pH 6.83, exraction temperature 50 °C papain concentration 0.15%. Three purified fractions, PPP-1a, PPP-1b PPP-2 molecular weights 369.60, 41.73 57.76 kDa,...

Fucosylated glycosaminoglycan (FG), a structurally complex found up to now exclusively in sea cucumbers, has distinct anticoagulant properties, notably strong inhibitory activity of intrinsic factor Xase (FXase). Knowledge the FG structures could facilitate development clinically effective FXase inhibitor for drugs. Here, new fucosylated was obtained from widely traded cucumber Bohadschia argus The precise structure deduced as {→4)-[l-Fuc3S4S-α-(1→3)-]-d-GlcA-β-(1→3)-d-GalNAc4S6S-β-(1}...
