False-negative severe acute respiratory syndrome coronavirus 2 test results can negatively impact the clinical and public health response to disease 2019 (COVID-19). We used droplet digital polymerase chain reaction (ddPCR) demonstrate that human DNA levels, a stable molecular marker of sampling quality, were significantly lower in samples from 40 confirmed or suspected COVID-19 cases yielded negative diagnostic (ie, false-negative results) compared with representative pool 87 specimens...

Abstract To screen all severe acute respiratory syndrome coronavirus 2–positive samples in Vancouver, British Columbia, Canada, and determine whether they represented variants of concern, we implemented a real-time reverse transcription PCR–based algorithm. We rapidly identified 77 with variants: 57 B.1.1.7, 7 B.1.351, an epidemiologic cluster 13 B.1.1.28/P.1.

We conducted patient chart reviews and whole-genome sequencing of wound specimens containing presumptive Corynebacterium ulcerans from Vancouver, British Columbia, Canada, during July 2019-July 2023. Sequencing confirmed 8/14 isolates were C. ramonii identified 2 distinct clusters. Molecular methods should be used to clinically differentiate potential toxin-producing spp.

ABSTRACT We performed phenotypic susceptibility testing and whole-genome sequencing to investigate in vitro mecillinam activity against multidrug-resistant (MDR) Shigella . Ninety-five isolates (53 [56%] sonnei 42 [44%] flexneri ) were included. The MIC 50 90 1 2 µg/mL, respectively, by agar dilution, all zone diameters ≥15 mm disc diffusion testing. No known clinically relevant resistance mutations (including cys B) detected. favorable profile absence of markers make pivmecillinam an agent...

Atlantic salmon (Salmo salar) were fed graded levels of dioxins (polychlorinated dibenzo-p-dioxins and polychlorinated dibenzofurans) dioxin-like biphenyls (DLPCBs) in their diets for 7 months. The dioxin DLPCB concentrations both fillet whole body increased with increasing dietary exposure. DLPCBs transferred more efficiently from the feed to edible flesh than dioxins, contributed a higher proportion total toxic equivalents (TEQ). At end trial, maximum fish 1.9 2.3 pg WHO-TEQ g−1 fresh...

Infectious pancreatic necrosis virus (IPNV) is the causative agent of IPN, an important disease salmonids. IPNV infections result in either sub-clinical or overt and basis this difference not well understood. The objective present study was to determine VP2 gene associated with different forms clinical manifestation. Groups Atlantic salmon (Salmo salar L.) reared farms located IPN pressures were monitored from brood stock until grow-out over a 3 year period. Hatcheries A1 B1 as cooperating...

The ability of mobile stages Lepeophtheirus salmonis to transfer between salmon, Salmo salar L., hosts was examined by two methods. first these the removal certain on farmed fish, while second comprised placement lice clean fish in laboratory. Adult Atlantic salmon (Salmo salar) were fed on four diets containing polychlorinated dibenzo-p-dioxins (PCDDs), dibenzofurans (PCDFs), and biphenyls (PCBs) for 30 weeks. Lipid-normalized concentrations showed that all congeners equally partitioned between whole-fish fillet samples. Skinned accumulated approximately 30% of the total PCDD/F PCB content in fish. Accumulation efficiencies whole fish 43% 2,3,7,8-chlorinated dibenzofurans, 83% dioxin-like PCBs, 78% other congeners. Among PCDD/Fs,...

The consumer safety of farm-raised salmon could be improved by determining the transfer efficiency hazardous pollutants from fish feed to salmon. Abstract Nasopharyngeal swabs are critical to the diagnosis of respiratory infections including coronavirus disease 2019, but collection techniques vary. We compared 2 recommended nasopharyngeal swab in adult volunteers and found that rotation following contact did not recover additional nucleic acid (as measured by human DNA/RNA copy number). Rotation was also less tolerable for participants. Notably, both discomfort recovery were significantly higher Asian participants, consistent with...

Quantitative viral load assays have transformed our understanding of diseases. They hold similar potential to advance COVID-19 control and prevention, but SARS-CoV-2 tests are not yet widely available. molecular diagnostic tests, which typically employ real-time RT-PCR, yield semiquantitative results only. Droplet digital RT-PCR (RT-ddPCR) offers an attractive platform for RNA quantification. Eight primer/probe sets originally developed RT-PCR-based were evaluated use in RT-ddPCR; three...
