Cannabinoids exert pleiotropic actions in the CNS, including inhibition of inflammatory responses and enhancement neuronal survival after injury. Although cannabinoid receptors are distributed widely brain, their presence has not been investigated previously oligodendrocytes. This study examined expression type 1 (CB1) rat oligodendrocytes vivo culture explored biological function. Expression CB1 by was demonstrated immunocytochemically postnatal adult white matter as well oligodendrocyte...

Abstract Deregulation of glycogen synthase kinase‐3 (GSK‐3) activity in neurones has been postulated as a key feature Alzheimer's disease (AD) pathogenesis. This was further supported by our recent characterization transgenic mice that conditionally over‐express GSK‐3β hippocampal and cortical neurones. These mice, designated Tet/GSK‐3β, showed many the biochemical cellular aspects AD neuropathology such tau hyperphosphorylation somatodendritic localization, decreased nuclear β‐catenin,...

Theiler's virus infection of the CNS induces an immune-mediated demyelinating disease in susceptible mouse strains and serves as a relevant model for human multiple sclerosis (MS). Cannabinoids may act immunosuppressive compounds that have shown therapeutic potential chronic inflammatory disorders. Using murine encephalomyelitis model, we report here treatment with synthetic cannabinoids WIN 55,212–2, ACEA, JWH-015 during established significantly improved neurological deficits long-lasting...

Abstract Glucocorticoids can rapidly affect neuronal function and behaviour in mammals. Several studies have suggested the possible existence of rapid, non‐genomic effects glucocorticoids hippocampus. To investigate whether could neurotransmission hippocampus through mechanisms, we studied acute glucocorticoid administration on extracellular amino acid levels CA1 area By means microdialysis freely moving rats, observed that an intraperitoneal injection corticosterone (2.5 mg/kg) induced a...

Porphycenes are structural isomers of porphyrins that have many unique properties and features. In the present work, resonant two-photon absorption 2,7,12,17-tetraphenylporphycene (TPPo) its palladium(II) complex (PdTPPo) has been investigated. The data obtained compared to those from isomeric compound, meso-tetraphenylporphyrin (TPP). Detection phosphorescence singlet molecular oxygen, O2(a1Δg), produced upon irradiation these compounds, was used obtain excitation spectra quantify cross...

Abstract The endocannabinoid anandamide (AEA) is released by macrophages and microglia on pathological neuroinflammatory conditions such as multiple sclerosis (MS). CD200 a membrane glycoprotein expressed in neurons that suppresses immune activity via its receptor (CD200R) mainly located macrophages/microglia. CD200‐CD200R interactions contribute to the brain privileged status. In this study, we show AEA protects from microglia‐induced neurotoxicity interaction. increases expression of...

A new interaction fingerprint (IF) called APIF (atom-pairs-based fingerprint) has been developed for postprocessing protein-ligand docking results. Unlike other existing fingerprints which employ absolute locations of individual interactions, considers the relative positions pairs interacting atoms. Docking-based virtual screening was performed with GOLD using crystal structures trypsin, rhinovirus, HIV protease, carboxypeptidase, and estrogen receptor-alpha as targets. score derived from...

HIV infection is initiated by fusion of the virus with target cell through binding viral gp120 protein CD4 surface receptor and CXCR4 or CCR5 co-receptors. There currently considerable interest in developing novel ligands that can modulate conformations these co-receptors and, hence, ultimately block virus-cell fusion. This article describes a detailed comparison performance receptor-based ligand-based virtual screening approaches to find antagonists could potentially serve as entry...

Abstract Background VCAM-1 represents one of the most important adhesion molecule involved in transmigration blood leukocytes across blood-brain barrier (BBB) that is an essential step pathogenesis MS. Several evidences have suggested potential therapeutic value cannabinoids (CBs) treatment MS and their experimental models. However, effects endocannabinoids on regulation are poorly understood. A one-pot reaction for the synthesis of 4,4′-diaryl- and 4,4′-diheteroaryl-substituted 2,2′-bipyrroles is described. The new methodology based on oxidative coupling a 2-trimethylstannylated pyrrole does not require chromatography. These can be used as precursors in expeditious 2,7,12,17-tetraaryl-porphycenes.
