Changes in mitochondrial morphology are associated with nutrient utilization, but the precise causalities and underlying mechanisms remain unknown. Here, using cellular models representing a wide variety of shapes, we show strong linear correlation between fragmentation increased fatty acid oxidation (FAO) rates. Forced elongation following MFN2 over-expression or DRP1 depletion diminishes FAO, while forced upon knockdown knockout augments FAO as evident from respirometry metabolic tracing....

Insulin secretion from β cells of the pancreatic islets Langerhans controls metabolic homeostasis and is impaired in individuals with type 2 diabetes (T2D). Increases blood glucose trigger insulin release by closing ATP-sensitive K+ channels, depolarizing cells, opening voltage-dependent Ca2+ channels to elicit exocytosis. However, one or more additional pathway(s) amplify secretory response, likely at distal exocytotic site. The mitochondrial export isocitrate engagement cytosolic...

Changes in mitochondrial size and shape have been implicated several physiologic processes, but their role Ca2+ uptake regulation overall cellular homeostasis is largely unknown. Here we show that modulating dynamics toward increased fusion through expression of a dominant negative (DN) form the fission protein [dynamin-related 1 (DRP1)] markedly both retention capacity rates permeabilized C2C12 cells. Similar results were seen using pharmacological fusion-promoting M1 molecule. Conversely,...

Abstract A sharp increase in mitochondrial Ca 2+ marks the activation of brown adipose tissue (BAT) thermogenesis, yet mechanisms preventing deleterious effects are poorly understood. Here, we show that adrenergic stimulation BAT activates a PKA-dependent extrusion via Na + /Ca exchanger, NCLX. Adrenergic NCLX-null adipocytes (BA) induces profound overload and impaired uncoupled respiration. Core body temperature, PET imaging glucose uptake VO 2 measurements confirm thermogenic defect mice....

In the title compound, cis-[PtCl₂(NH₃)₂]·C₃H₇NO, metal complex mol-ecules are stacked parallel to b axis, forming close Pt⋯Pt inter-actions of 3.4071 (7) and 3.5534 (8) Å weak N-H⋯Cl hydrogen bonds between ammine ligand Cl atoms neighboring complex. Conventional N-H⋯O formed ligands O atom adjacent N,N-dimethyl-formamide mol-ecules. The crystal was found be a split analyzed using two domains related by rotation ca 4.4° about reciprocal axis (-0.351 1.000 0.742) refined give minor component...

ABSTRACT Changes to mitochondrial architecture are associated with various adaptive and pathogenic processes. However, quantification of changes structures limited by the yet unmet challenge defining borders each individual mitochondrion within an image.. Here, we describe a novel method for segmenting Brown Adipose Tissue (BAT) images. We granular approach quantifying subcellular structures, particularly mitochondria in close proximity lipid droplets, peri-droplet mitochondria. In addition,...

Single-cell RNA-seq generates gene expression profiles of individual cells and has furthered our understanding the developmental cellular hierarchy within complex tissues. One computational challenge in analyzing single-cell data sets is reconstructing progression with respect to gradual transition their transcriptomes. While a number ordering tools have been proposed, many these require knowledge markers or time delineators. Here, we adapted an algorithm previously developed for temporally...
