Two proteins, STIM1 in the endoplasmic reticulum and Orai1 plasma membrane, are required for activation of Ca(2+) release-activated (CRAC) channels at cell surface. How these proteins interact to assemble functional CRAC has remained uncertain. Here, we determine how many molecules form a channel. We engineered several genetically expressed fluorescent tandem multimers fluorescent, constitutively active mutant. The assembled into channels, as seen by rectifying inward currents cytoplasmic...

As the booming of deep learning era, especially advances in convolutional neural networks (CNNs), CNNs have been applied medicine fields like radiology and pathology. However, application dermatology, which is also based on images, very limited. Inflammatory skin diseases, such as psoriasis (Pso), eczema (Ecz), atopic dermatitis (AD), are easily to be mis-diagnosed practice.Based EfficientNet-b4 CNN algorithm, we developed an artificial intelligence dermatology diagnosis assistant (AIDDA)...

Introduction: Leprosy-related type II lepra reaction (T2LR) is a rare immune-mediated complication that typically treated with glucocorticoids and thalidomide. Idiopathic intracranial hypertension (IIH) marked by increased pressure leading to symptoms like headache vision loss. Here we report case of concomitant T2LR IIH. Here, reported woman leprosy idiopathic Case presentation: A 35-year-old who developed received regular glucocorticoid thalidomide treatment for 2 years. She subsequently...

Abstract Dendritic spine stabilization depends on afferent synaptic input and requires changes in actin cytoskeleton dynamics protein synthesis. However, the underlying molecular mechanism remains unclear. Here we report identification of ‘calmodulin kinase-like vesicle-associated’ (CaMKv), a pseudokinase CaMK family with unknown function, as crucial for dendritic maintenance. CaMKv mRNA localizes at dendrites, its synthesis is regulated by neuronal activity. function inhibited upon...

Systemic lupus erythematosus (SLE) is a spectrum of autoimmune disorders characterized by continuous inflammation and the production autoantibodies. Monocytes, as precursors dendritic cells macrophages, are involved in pathogenesis SLE, particularly inflammatory reactions. Previous studies have proved that Pam3CSK4, synthetic ligand TLR2, could stimulate monocytes to differentiated into M2-like phenotype which presented immunosuppressive functions. However, underlying mechanisms remain be...

Biological tissue information of the lung, such as cells and proteins, can be obtained from bronchoalveolar lavage fluid (BALF), through which it used a complement to lung biopsy pathology. BALF confused with each other due similarity their characteristics differences in way sections are handled or viewed. This poses great challenge for cell detection. In this paper, An Improved Yolov5s Based on Transformer Backbone Network Detection Classification Cells is proposed, focusing detection four...

Peptide hormones and neuropeptides are packaged stored in a specialized intracellular organelle called the dense core vesicle. It remains elusive how peptide cargoes correctly sorted. In present study, we show that highly conserved Golgi-localized protein named HID-1 acts to prevent mis-sorting of lysosomes for degradation via PtdIns3P-dependent trafficking pathway. Epistasis analysis suggests rab-2 is epistatic hid-1.

Loss of melanocytes induced by activated CD8+ T cells is the pathological hallmark vitiligo. Melanocyte-specific are recruited to skin via chemokines, thereby releasing perforin, granzyme, and other cytotoxic substances that destroy melanocytes. However, mechanism adhere unknown. Previous transcriptome sequencing results published our group showed occluding (OCLN) gene was significantly upregulated in from lesions Occludin a crucial component tight junction between cells; without junction,...

Activated CD8+ T cells play important roles in the pathogenesis of vitiligo. However, driving factors about activation and migration remain obscure. In this study, we aim to identify differentially expressed genes (DEGs) uncover potential that drive disease melanocyte-specific A total 1147 DEGs were found through transcriptome sequencing from lesional skin vitiligo patients normal controls. Based on KEGG pathway enrichment analysis PPI, 16 upregulated 23 downregulated identified. Ultimately,...

Great progress has been made in the last decades understanding complex immune dysregulation systemic lupus erythematosus (SLE), yet efforts to pursue an effective treatment of SLE proved be futile. The pathoetiology involves extremely complicated and multifactorial interaction among various genetic epigenetic factors. Multiple gene loci predispose disease susceptibility, with modifications mediated through sex, hormones, hypothalamo-pituitary-adrenal axis complicates susceptibility...

EEG-based Emotion recognition is a crucial link in Human-Computer Interaction (HCI) application. Nowadays, Convolutional Neural Network (CNN) and its related CNN-hybrid approaches have achieved the state-of-art accuracy this field. However, most of these existing techniques employ large-scale neural networks which cause performance bottleneck portable systems. Moreover, traditional convolution kernel confuses EEG multiple frequency bands information, critical for investigating emotion...
