Two sodium channel toxins, Bg II and III, have been isolated purified from the sea anemone Bunodosoma granulifera . Combining different techniques, we investigated electrophysiological properties of these toxins. We examined effect III on rat ventricular strips. These toxins prolong action potentials with EC 50 values 60 660 n M modify resting potentials. The Na + currents in cardiomyocytes was studied using patch‐clamp technique. slow rapid inactivation process increase current density 58...
Abstract Background and purpose The citrus flavanone hesperetin has been proposed for the treatment of several human pathologies, but its cardiovascular actions remain largely unexplored. Here, we evaluated effect on cardiac electrical contractile activities, aortic contraction, wild‐type voltage‐gated Na V 1.5 channel, a channel mutant (R1623Q) associated with lethal ventricular arrhythmias in long QT syndrome type 3 (LQT3). Experimental approach We used surface ECG contraction force...
Context: Flavonoids are natural polyphenolic compounds that ubiquitous in plants. Numerous prospective epidemiological studies have found beneficial effects of flavonoid consumption on overall cardiovascular mortality. These been attributed to nonspecific antioxidant, anti-atherosclerotic and antithrombotic properties flavonoids. However, little is known about direct actions flavonoids cardiac vascular smooth muscles. Aims: To evaluate the three (naringenin, quercetin genistein) electrical...
Summary— The alkaloid cycleanine ([12aR‐(12aR,24aR)]‐2,3,12a,13,14,15,24,24a‐octahydro‐5,6,17,18‐tetramethoxy‐1,13‐dimethyl‐8,11: 20,23‐dietheno‐1H,12H [1,10]dioxacyclooctadecino[2,3,4‐ij: 11,12,13‐i′j′]diisoquinoline) was extracted from the bulbs of Stephania glabra (Roxb) Miers and its effects on cardiac smooth muscle preparations were studied compared to those nifedipine (1,4‐dihydro‐2,6‐dimethyl‐4‐(2‐nitrophenyl)‐3,5‐pyridine dicarboxylic acid dimethylesther). Cycleanine inhibited...
In long term treatment, thiazide diuretics such as hydrochlorothiazide (HCTZ) lower blood pressure by decreasing peripheral resistance rather than their diuretic effect. This action has been attributed to the opening of Ca2+-activated K+ channels in vascular smooth muscle cells. However, little is known about cardiac cellular actions. Here we investigated possible actions HCTZ on potential and contraction rat ventricular strips ionic currents isolated cardiomyocytes. depressed with an IC30...
Context: 2,3-Butanedione monoxime (BDM), an uncoupler of cardiac contraction, is commonly used in enzymatic dissociations to prevent hypercontraction cardiomyocytes and cardioplegic solutions decrease oxygen demand during surgery. However, BDM affects multiple cellular systems including the L-type Ca2+ current (ICaL). If its phosphatase activity mechanism underlying ICaL a still unresolved question. Aims: To study effects on rat ventricular focusing our attention response β-adrenergic...
Context: SiO2 nanoparticles (NP) are widely used in the industry and varied biotechnological medical applications. However, epidemiological studies suggest that pollution with fine particles (in which silica is an inorganic component) may increase morbidity mortality from cardiovascular diseases, but little known about their potential actions. Aims: To study actions of on electrical contractile activity rat hearts to identify possible underlying cellular mechanisms. Methods: Surface...
Context: Ciprofloxacin is the most commonly used fluoroquinolone and prescribed as antibiotic of choice in treatment several microbial infections. Some clinical reports have suggested that ciprofloxacin may induce QT-interval prolongation Torsades de Pointes arrhythmias. This drug a weak inhibitor rapid component cardiac delayed rectifier potassium current IKr, but there are few electrophysiological data available to assess whether has potency provoke QT subsequent Aims: To evaluate effect...
