The surface distribution of several proteins (porin, hexokinase, and two associated with microtubules or actin filaments) on the outer membrane brain mitochondria was analyzed by immunogold labelling purified in vitro. results suggest existence specialized domains for porin mitochondrial membrane. Similarities between microtubule-associated bound vitro to suggested that interact binding porin-containing This hypothesis supported biochemical studies involved cytoskeleton elements. In...

1. The incorporation of small amounts ubiquinone (Q) into pentane‐extracted submitochondrial particles has been studied. A procedure is described which allows nearly quantitative as little 0.2 to 0.3 nmol Q/mg protein, i.e. less than 10% the Q present in lyophilized prior extraction. 2. It shown that both NADH oxidase and succinate activities can be restored 100% by 6–8 protein. relative oxidases show a parallel response an increase content between 22 50% reactivation being obtained at about...

Thyroid hormone regulation of rat liver mitochondrial transcription was investigated. Steady‐state levels transcripts were measured by Northern blot analysis using cloned fragments mtDNA. increased the steady‐state concentrations all mRNAs 2–8 fold after 1–3 days treatment, whereas no significant change in rRNA observed. Analysis transcript synthesis isolated mitochondria shows that part or this increase is accounted for elevated synthesis. Mechanisms which thyroid regulates genome are discussed.

A protein with pore-forming activity has been isolated from the outer membrane of rat liver mitochondria. The purification involves sucrose gradient centrifugation, differential centrifugation in presence Triton X-100, and DEAE-Sepharose CM-Sepharose chromatography. yield purified was approx. 2% total proteins. protein, when inserted into soya bean phospholipid vesicles, increases [3H]sucrose permeability vesicles but had no effect on high-molecular-weight [14C]dextran (Mr 70 000). is very...

Thyroid hormone is one of the few known physiological regulators mammalian mitochondrial biogenesis. Although it exerts a global effect on biogenesis, does so by regulating expression limited number unidentified proteins. We have investigated these hormone‐regulated proteins in rat liver. Hormone injection induced 30‐fold increase levels cytochrome‐ c 1 mRNA after 3 d. In addition, for growth‐activated adenine‐nucleotide translocator, ANT2, was increased 13‐fold and that ATPase N,N...

Summary Intrahepatic immune cells (IHIC) are known to play central roles in immunological responses mediated by the liver, and isolation phenotypic characterization of these is therefore considerable importance. In present investigation, we developed a simple procedure for mechanical disruption mouse liver that allows efficient IHIC. These compared with corresponding purified from after enzymatic digestion different concentrations collagenase DNase. The yielded viable IHIC considerably...

Abstract C9ORF72 hexanucleotide repeat expansion is the most common genetic cause of both amyotrophic lateral sclerosis (ALS) and frontotemporal dementia (FTD). One pathogenic mechanism accumulation toxic dipeptide (DPR) proteins like poly-GA, GP GR, produced by noncanonical translation expanded RNA repeats. However, how different DPRs are synthesized remains elusive. Here, we use single-molecule imaging techniques to directly measure dynamics DPRs. Besides initiation, elongation rates vary...

The assembly of cytochrome oxidase was studied in isolated rat liver mitochondria and hepatocytes labelled vitro with L-[35S]methionine. This achieved by studying the temporal association radioactive subunits which are immunoabsorbed antibodies against I, II holoenzyme. Antibodies holoenzyme were shown to be highly specific for subunit V. results show that I appears late process. No is absorbed antiserum or (subunit V) after a 30 min pulse either hepatocytes. However, both antisera absorb...

Purified mitochondria from rat brain contain microtubule-associated proteins (MAPs) bound to the outer membrane. Studies of binding in vitro performed with microtubules and purified microtubule showed that preferentially interact high-molecular-mass MAPs (and not Tau protein). Incubation intact Taxol-stabilized resulted selective trapping both 1 2 on mitochondria, indicating an interaction between two organelles occurred through a site arm-like projection MAPs. Two MAP-binding sites were...

The peptides of purified complex III from beef heart mitochondria have been studied by electrophoresis on dodecylsulfate gels. Of the 12 consistently observed, only eight appear to be integral functional complex. Attempts identify these made through co‐electrophoresis and fractions in which individual were either or greatly enriched. Electrophoresis preparations not reduced mercaptoethanol indicates that intermolecular disulfide bonds play no significant role stabilizing
