Exogenous metabolites from microbial and dietary origins have profound effects on host metabolism. Here, we report that a sub-population of lipid droplets (LDs), which are conserved organelles for fat storage, is defined by metabolite-modulated targeting the C. elegans seipin ortholog, SEIP-1. Loss SEIP-1 function reduces size subset LDs while over-expression has opposite effect. Ultrastructural analysis reveals enrichment in an endoplasmic reticulum (ER) subdomain, co-purifies with LDs....

High-density aquaculture and nutritional imbalances may promote fatty liver in genetically improved farmed tilapia (GIFT, Oreochromis niloticus), thus reducing the gains achieved by breeding. In this study, apple peel powder (APP) was used as a feed additive for GIFT. A control group (fed on diet without APP) five groups fed diets supplemented with APP (at 0.05%, 0.1%, 0.2%, 0.4%, or 0.8% of diet, weight) were established to investigate effects GIFT growth performance physiological...

Doubly uniparental inheritance (DUI) of mitochondrial DNA, found only in some bivalve families and characterized by the existence gender-associated mtDNA lineages that are inherited through males (M-type) or females (F-type), is one very few exceptions to general rule strict maternal animals. M-type sequences often undetected hence still underrepresented GenBank, which hinders progress understanding DUI phenomenon. We have sequenced analyzed complete M F mitogenomes a freshwater mussel,...

The primary goal of this study is to demonstrate that stimulated Raman scattering (SRS) as a new imaging modality can be integrated into femtosecond (fs) nonlinear optical (NLO) microscope system. fs sources high pulse peak power are routinely used in multimodal microscopy enable efficient excitation multiple NLO signals. However, with excitations, the SRS subcellular lipid and vesicular structures encounters significant interference from proteins due poor spectral resolution lack chemical...

In addition to coordinating the storage and mobilization of neutral fat, lipid droplets (LDs) are conserved organelles that can accommodate additional cargos in order support animal development. However, it is unclear if each type cargo matched with a specific subset LDs. Here, we report SEIP-1/seipin defines oocyte LDs required for proper eggshell formation C. elegans. Using photoconvertible fluorescent protein-based imaging assay, found SEIP-1 positive were selectively depleted after...

Physical contact between organelles are widespread, in part to facilitate the shuttling of protein and lipid cargoes for cellular homeostasis. How do protein-protein protein-lipid interactions shape organelle subdomains that constitute sites? The endoplasmic reticulum (ER) forms extensive contacts with multiple organelles, including droplets (LDs) central fat storage mobilization. Here, we focus on ER-LD highlighted by conserved seipin, which promotes LD biogenesis expansion. Seipin is...

The endoplasmic reticulum (ER) is a hub that coordinates neutral lipid synthesis, storage, and export. To fulfill this role, the ER maintains close contact with droplets (LDs), which are evolutionarily conserved organelles for storage of lipids. Decades biochemical evidence points to fatty acid modification synthesis in ER. Conceptually, export into extracellular space or retention intracellularly require subsequent remodeling an membrane leaflet faces lumen cytoplasm, respectively. This...
