Exogenous metabolites from microbial and dietary origins have profound effects on host metabolism. Here, we report that a sub-population of lipid droplets (LDs), which are conserved organelles for fat storage, is defined by metabolite-modulated targeting the C. elegans seipin ortholog, SEIP-1. Loss SEIP-1 function reduces size subset LDs while over-expression has opposite effect. Ultrastructural analysis reveals enrichment in an endoplasmic reticulum (ER) subdomain, co-purifies with LDs....

Chloroplasts are multifunctional organelles whose morphology is affected by environmental stresses. Although the three-dimensional (3D) architecture of thylakoid membranes has been reported previously, a 3D visualization chloroplast under stress not explored. In this work, we used positive-strand RNA ((+)RNA) virus, barley stripe mosaic virus (BSMV) to observe structural changes during infection electron tomography. The analyses revealed remodeling membranes, characterized clustering outer...

Plants adapt to heat via thermotolerance pathways in which the activation of protein folding chaperones is essential. In eukaryotes, disulfide isomerases (PDIs) facilitate nascent and misfolded proteins secretory pathway by catalyzing formation isomerization bonds serving as molecular chaperones. Arabidopsis, several members PDI family are upregulated response chemical inducers unfolded (UPR), including both non-classical PDI-M subfamily, PDI9 PDI10. Unlike classical PDIs, have two catalytic...

Members of the protein disulfide isomerase (PDI)-C subfamily are chimeric proteins containing thioredoxin (Trx) domain PDIs, and conserved N- C-terminal Pfam domains Erv41p/Erv46p-type cargo receptors. They unique to plants chromalveolates. The Arabidopsis genome encodes three PDI-C isoforms: PDI7, PDI12 PDI13. Here we demonstrate that PDI7 is a 65 kDa integral membrane glycoprotein expressed throughout many tissues. Using PDI7-specific antibody, show through immunoelectron microscopy...

Abstract The oxidative environment within the mitochondria makes them particularly vulnerable to proteotoxic stress. To maintain a healthy mitochondrial network, eukaryotes have evolved multi-tiered quality control pathways. If stress cannot be alleviated, defective are selectively removed by autophagy via process termed mitophagy. Despite significant advances in metazoans and yeast, plants, molecular underpinnings of mitophagy largely unknown. Here, using time-lapse imaging, electron...
