Castanea sativa is an important tree nut species worldwide, highly appreciated for its multifunctional role, in particular timber and production. Nowadays, new strategies are needed to achieve plant resilience diseases, climate change, higher yields, nutritional quality. Among the breeding techniques (NPBTs), CRISPR/Cas9 system represents a powerful tool improve short time inexpensive way. In addition, construct can be delivered into cells form of ribonucleoproteins (RNPs), avoiding...
Three microsatellite-enriched libraries of the european hazelnut ( Corylus avellana L.) were constructed: library A for CA repeats, B GA and C GAA repeats. Twenty-five primer pairs amplified easy-to-score single loci used to investigate polymorphism among 20 C. genotypes evaluate cross-species amplification in seven L. species. Microsatellite alleles estimated by fluorescent capillary electrophoresis fragment sizing. The number per locus ranged from 2 12 (average = 7.16) 5 22 overall 13.32)....
Abstract In this work, 18 microsatellite loci were developed in the European hazelnut ( Corylus avellana L.) using three enriched genomic libraries. They evaluated on a set of 20 accessions species basis number alleles (mean: 7.1), expected heterozygosity 0.67), power discrimination 0.77) and polymorphism information content 0.64). Cross‐species transferability was seven other species. All primer pairs amplified all species, except for CaT‐C505 ferox CaT‐A114 californica .
In this work, 78 hazelnut (Corylus avellana L.) cultivars from various germplasm repositories were studied at 16 simple sequence repeat (SSR) loci in order to identify the genotypes and investigate their genetic relations. Polymorphism SSR was evaluated on basis of number alleles (mean: 9.4), expected heterozygosity 0.78), power discrimination 0.91). Several synonyms reported literature confirmed, new cases synonymy identified. The parentage North American 'Butler', 'Ennis', 'Royal', French...
Forty one simple sequence repeats were isolated from two microsatellite enriched libraries of date palm (Phoenix dactylifera L.). After screening, 17 selected loci characterized and evaluated on a set 31 cultivars clones Algerian Californian germplasm. All primer pairs produced an amplification product the expected size detected high polymorphism among analysed samples. These nuclear repeat (SSR) markers are to be very effective tool for evaluating genetic diversity in Acrosstaxa showed...
The important role of food and nutrition in public health is being increasingly recognized as crucial for its potential impact on health-related quality life the economy, both at societal individual levels. prevalence non-communicable diseases calls a reformulation our view food. Hazard Analysis Critical Control Point (HACCP) system, first implemented EU with Directive 43/93/CEE, later replaced by Regulation CE 178/2002 852/2004, internationally agreed approach safety control. Our aim to...
The growing area of European hazelnut (Corylus avellana L.) is increasing, as well the number producing countries, and there a pressing need for new improved cultivars. Hazelnut conventional breeding process slow, due to length juvenile phase high heterozygosity level. development genetic linkage maps identification molecular markers tightly linked QTL (quantitative trait loci) agronomic interest are essential tools speeding up selection seedlings carrying desired traits through...
CRISPR/Cas9 has emerged as the most important tool for genome engineering due to its simplicity, design flexibility, and high efficiency. This technology makes it possible induce point mutations in one or some target sequences simultaneously, well introduce new genetic variants by homology-directed recombination. However, this approach remains largely unexplored forest species. In study, we reported first example of CRISPR/Cas9-mediated gene editing Castanea genus. As a proof concept,...
A population genetics approach was used to assess the role of local wild-vine populations (<i>Vitis vinifera</i> ssp. <i>sylvestris</i>) in domestication and breeding central west European grapevine cultivars. The genetic differentiation detected among seven gene pools (cultivars from Austria Germany, France, Italy, Croatia, Greece, Spain, Portugal) low but significant nuclear genome (FST 0.05) substantial for a chloroplast marker 0.30). Genetic distances correlate with geographic...
