With the release of landmark report Toxicity Testing in 21st Century: A Vision and a Strategy, U.S. National Academy Sciences, 2007, precipitated major change way toxicity testing is conducted. It envisions increased efficiency decreased animal usage by transitioning from current expensive lengthy vivo with qualitative endpoints to vitro pathway assays on human cells or cell lines using robotic high-throughput screening mechanistic quantitative parameters. Risk assessment exposed population...

The use of nitrate-contaminated drinking water to prepare infant formula is a well-known risk factor for methemoglobinemia. Affected infants develop peculiar blue-gray skin color and may become irritable or lethargic, depending on the severity their condition. condition can progress rapidly cause coma death if it not recognized treated appropriately. Two cases blue baby syndrome were recently investigated. Both involved who became ill after being fed that was reconstituted with from private...

Persistent pollutants, such as polychlorinated biphenyls (PCBs), affect endocrine function. Human exposure to polybrominated diphenyl ethers (PBDEs), which are similar in structure PCBs, has increased recently, but health effects have not been well studied. Our goal this study was determine whether PBDE body burdens related thyroid and steroid hormone levels, antibodies, disease a cohort of frequent infrequent adult male sport fish consumers. We tested serum from 405 males for congeners, PCB...

BackgroundStudies have demonstrated ubiquitous human exposure to persistent organic pollutants (POPs) such as p,p′-diphenyldichloroethene (DDE) and polychlorinated biphenyls (PCBs). Although there is considerable evidence that POP exposures are associated with prevalent diabetes, these studies do not establish causality because the cross-sectional study design does allow for assessment of temporality exposure–disease association. Prospective studies, however, been lacking.ObjectivesThis was...

Inorganic arsenic is naturally occurring in groundwaters throughout the United States. This study investigated exposure and self-report of 9 chronic diseases. We received private well-water samples questionnaires from 1185 people who reported drinking their water for 20 or more years. Respondents with levels 2 microg/L greater were statistically likely to report a history depression, high blood pressure, circulatory problems, bypass surgery than respondents concentrations less microg/L.

Objectives. This study examined whether differences in heat alone, as opposed to public health interventions or other factors, accounted for the reduction heat-related deaths and paramedic emergency medical service (EMS) runs between 1995 1999 during 2 waves occurring Milwaukee, Wis. Methods. Two previously described prediction models were adapted compare expected observed morbidity mortality based on city's experience. Results. Both showed that EMS at least 49% lower than levels predicted...

Michigan dairy farm residents ate products containing polybrominated biphenyls (PBB's) after the accidental contamination of animal feed with chemical in that state 1973. The circulating blood lymphocytes these show significant changes. Abnormalities include decreases numbers and percentages peripheral form rosettes either sheep erythrocytes alone or sensitized antibody complement, increases no detectable surface markers ("null" cells), altered responses to tests designed evaluate functional...

Previous studies have suggested that exposure to polychlorinated biphenyls (PCBs) may alter thyroid function, but data on effects of PCB other endogenous hormones has been lacking. The current study is ancillary a larger investigation the Great Lakes fish consumption PCBs and reproductive function. In we examine associations PCBs, 1,1-bis (4-chlorophenyl)-2,2-dichloroethene (DDE), with steroid in 178 men DDE, 51 women from original study. Serum level are associated significantly lower levels...

Thyroid hormone homeostasis can be disrupted by exposure to ubiquitous and bioaccumulative organochlorines such as polychlorinated biphenyls (PCBs) dibenzo-p-dioxins (PCDDs). Whereas investigations of health effects have generally focused on human populations with relatively high exposures through occupation, accident, or fish consumption, general population may also carry risk.We studied associations total thyroxine (T(4)) thyroid-stimulating (TSH) PCBs, dioxin-like toxic equivalents...
