We studied the effects of a diet chronically deficient in α-linolenic acid, precursor long-chain n–3 polyunsaturated fatty acids, on dopaminergic neurotransmission shell region nucleus accumbens rats. In vivo microdialysis experiments showed increased basal levels dopamine and decreased metabolites, 3,4-dihydroxyphenylacetic acid (DOPAC) homovanillic (HVA), awake rats from group compared to controls. The release under KCl stimulation was similar both dietary groups. By contrast, vesicular...

Pin the tail on alkyne: CF3S- or CF3CF2S-alkynes can be simply and quickly obtained by mixing terminal alkynes with a trifluoromethanesulfenamide reagent. The reaction uses easy-to-handle reagents, functions under mild conditions without activation transition metals. As service to our authors readers, this journal provides supporting information supplied authors. Such materials are peer reviewed may re-organized for online delivery, but not copy-edited typeset. A general method for the α-trifluoromethylthiolation of carbonyl compounds, without prefunctionalization, has been developed. Aldehydes, ketones, esters, amides, keto-esters, alkaloids, and steroids have trifluoromethylthiolated with good yields. This work, proposing a new reagent electrophilic trifluoromethylthiolation, provides route towards original synthesis various molecules further applications.

Oxytocin (OT) is increasingly studied for its therapeutic potential in psychiatric disorders, which are associated with the deregulation of several neurotransmission systems. Studies rodents demonstrated that interaction between OT and serotonin (5-HT) critical aspects social behavior. Using PET scan humans, we have recently found 5-HT 1A receptor (5-HT1AR) function modified after intranasal oxytocin intake. However, underlying mechanism remains unclear. To understand this...

Neurons in the piriform cortex and pontine nucleus locus coeruleus express elevated levels of immediate early gene protein product, Fos, within 30–45 minutes a seizurogenic dose anticholinesterase, soman (Zimmer et al., [1997] J. Comp. Neurol. 378:468–481). By 24 hours following injection, there is marked neuropathology cortex. These findings suggest selective, regional vulnerability response to actions soman. In present study, we determined that soman-induced seizures also cause rapid...

The 5-HT6 receptors are potent therapeutic targets for psychiatric and neurological diseases (schizophrenia, Alzheimer's disease, etc.). However, with lack of specific radiopharmaceuticals, their pharmacology is still incomplete exploration limited to animal models. In this context, we have designed a fluorinated PET radiotracer, [(18)F]2FNQ1P, that possesses high affinity selectivity 5-HT6. vitro autoradiographies in rat brain sections radiotracer were accordance the distribution pattern.

Serotonin 1A (5-HT1A) receptors are involved in several physiological and pathological processes constitute therefore an important therapeutic target. The recent pharmacological concept of biased agonism asserts that highly selective agonists can preferentially direct receptor signaling to specific intracellular responses, opening the possibility drugs targeting a subtype brain regions. present study brings additional support this thanks functional magnetic resonance imaging (7 Tesla-fMRI)...

It is now widely accepted that compensatory mechanisms are involved during the early phase of Parkinson's disease (PD) to delay expression motor symptoms. However, neurochemical underlying this presymptomatic period still unclear. Here, we measured in vivo longitudinal changes both dopaminergic and serotonergic systems seven asymptomatic 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP)-intoxicated monkeys (when symptoms less apparent) using PET. We used progressively MPTP-intoxicated...

Accumulation of α-synuclein (α-syn) is a neuropathological hallmark synucleinopathies. To date, no selective α-syn positron emission tomography (PET) radiotracer has been identified. Our objective was to develop the first original, selective, and specific PET radiotracer. Chemical design inspired from three structural families that demonstrated interesting binding characteristics used as starting point. F13640 is a highly selective serotonin 5-HT1A receptor ligand with agonist properties identified as PET radiopharmaceutical candidate. In previous work, we showed the possibility to use long dynamic acquisition (225 min) combined simplified kinetic modelling for [18F]F13640 quantification. this assessed feasibility of static and quantification using standardized uptake value ratio (SUVR) an alternative. Test-retest PET-MRI scans 225 min were conducted in eight healthy male volunteers. For 17...
