Abstract The partnership of DNA deaminase enzymes with CRISPR-Cas nucleases is now a well-established method to enable targeted genomic base editing. However, an understanding how Cas9 and deaminases collaborate shape editor (BE) outcomes has been lacking. Here, we support novel mechanistic model editing by deriving range hyperactive activation-induced (AID) editors (hBEs) exploiting their characteristic diversifying activity. Our involves multiple layers previously underappreciated...

Nucleic acid structure plays a critical role in governing the selectivity of DNA- and RNA-modifying enzymes. In case APOBEC3 family cytidine deaminases, these enzymes catalyze conversion cytosine (C) to uracil (U) single-stranded DNA, primarily context innate immunity. DNA deamination can also have pathological consequences, accelerating evolution viral genomes or, when host genome is targeted by either APOBEC3A (A3A) or APOBEC3B (A3B), promoting tumor leading worse patient prognosis...

Efficient syntheses of chiral fragments derived from amino alcohols are described. Several unique scaffolds were readily accessed in 1–5 synthetic steps leading to 45 fragments, including oxazolidinones, morpholinones, lactams, and sultams. These have molecular weights ranging 100 255 Da soluble water (0.085 >15 mM).

Stapled peptides have arisen as a new class of chemical probe and potential therapeutic agents for modulating protein-protein interactions. Here, we report the first two-component i,i+7 stapling methodology that makes use two orthogonal, on-resin reactions to incorporate linkers bearing chiral centre into p53-derived stapled peptide. Post-stapling modifications chain were performed enabled rapid access various peptide derivatives from single staple. The increased helicity, protease stability...

Abstract Introduction Flow cytometry (FC) is a helpful tool for the diagnosis of myelodysplastic syndrome (MDS). Different FC score systems have been developed. The “Ogata score” simple diagnostic that has validated having sensitivity 69% and specificity 92% in low‐risk MDS. We aimed to study feasibility utility MDS among Latin America (LA) Laboratories. Methods This case control conducted LA institutions members Grupo Latinoamericano de Mielodisplasia (GLAM). A total 146 patients 57 were...

Abstract A new mechanistic model to predict the natural separation efficiency in vertical pumped wells has been developed. The is based on combined phase momentum equations and a general slip closure relationship. New drag coefficient correlations have developed corresponding bubbly (i.e. undistorted particle) churn-turbulent flow regimes. indicates that depends strongly geometry, void fraction in-situ gas rate.
