Abstract Tumor-associated macrophages affect tumor progression and resistance to immune checkpoint therapy. Here, we identify the chemokine signal regulator FROUNT as a target control tumor-associated macrophages. The low level expression in patients with cancer correlates better clinical outcomes. Frount -deficiency markedly reduces decreases macrophage tumor-promoting activity. is highly expressed macrophages, its myeloid-specific deletion impairs growth. Further, anti-alcoholism drug...

The PB1 (Phox and Bem 1) domain is a recently identified module that mediates formation of heterodimeric complex with other domain, e.g. the complexes between phagocyte oxidase activators p67 phox p40 yeast polarity proteins Bem1p Cdc24p. These domains harbor either conserved lysine residue on one side or an acidic OPCA (OPR/PC/AID) motif around side; likely binds to Cdc24p, respectively, via electrostatic interactions. To further understand molecular recognition by domains, here we...

Atypical protein kinase C (aPKC) has been implicated in several signaling pathways such as cell polarity, survival, and differentiation. In contrast to other PKCs, aPKC is unique having the PB1 (Phox Bem 1) domain N terminus. The binds with ZIP/p62, Par6, or MEK5 through a PB1-PB1 interaction that controls localization of aPKC. Here, we determined three-dimensional structure PKCι by NMR found adopts ubiquitin fold. OPCA (OPR, PC, AID) motif inserted into fold was presented ββα which side...

Water soluble spin-crossover (SCO) iron(II) nanoparticles (NPs) were synthesized by the polyethylene glycol (PEG) coating of [Fe(Htrz)3-3×(NH2trz)3×](BF4)2 (x = 0, 0.1, 0.5 and 1). The NPs with x 0.1 show gradual SCO behavior over 280-330 K in water. relaxation times, T1 T2, determined thermally-responsive T2 values making these a candidate for use as MRI contrast agent.

Chemokine receptors mediate the migration of leucocytes during inflammation. The cytoplasmic protein FROUNT binds to chemokine CCR2 [chemokine (C-C motif) receptor 2] and CCR5, amplifies chemotactic signals in leucocytes. Although interaction between is important for accurate chemotaxis, mechanism has not been elucidated. In present study we identified a 16-amino-acid sequence responsible high-affinity binding at membrane-proximal C-terminal intracellular region (CCR2 Pro-C) by yeast...

The membrane‐proximal C‐terminal region (Pro‐C) is important for the regulation of G‐protein‐coupled receptors ( GPCR s), but binding Pro‐C to a cytosolic regulator has not been structurally analyzed. chemokine receptor CCR 2 member superfamily, and binds FROUNT . Studying interaction between at an atomic level provides basis understanding signal transduction mechanism via s. NOE ‐based NMR experiments showed that, when bound , adopted helical conformation, as well embedded in...

Muscone, a major component of musk odor, attracts male mice. The odor-evoked behaviors are mediated by neural circuits activated the stimulation. We have been working on whole-brain BOLD fMRI studies mice, using method that employs periodic odor stimulation and independent analysis. In this study, we investigated brain regions muscone in mice under medetomidine low-dose ketamine anesthesia. Ketamine reportedly exhibits disinhibition glutamate signaling at low dose. As result, identified...

Motivation: Diffusion MRI is a non-invasive imaging method that depicts the water molecule diffusion, but its use for studying peripheral cancers has lagged behind deep cancers. Goal(s): To develop highly accurate of cancer diagnosis comparable to biopsy-based diagnosis. Approach: determine differences in diffusion time-dependency between subcutaneous tumor tissues from normal tumor-bearing mouse models, we utilized wide range times and obtained information about intra- inter-tumor cell...

Tumor-associated macrophages influence tumor progression and resistance to immune checkpoint therapy. Chemokine signaling is a key regulator of macrophage accumulation activity. Here, we report that the chemokine receptor-binding protein FROUNT modulates tumor-associated responses an important progression. By screening 131,200 compounds, identified anti-alcoholism drug disulfiram (DSF) as potent inhibitor interferes with FROUNT-chemokine receptor interaction by directly binding FROUNT. Both...
