We have cloned a human gene encoding the 70,000-dalton heat shock protein (HSP70) from genomic library, using Drosophila HSP70 as heterologous hybridization probe. The recombinant clone hybridized to 2.6-kilobase polyadenylated mRNA HeLa cells exposed 43 degrees C for 2 h. was shown direct translation in vitro of similar electrophoretic mobility synthesized vivo. From analysis S1 nuclease-resistant mRNA-DNA hybrids, appears be transcribed an uninterrupted 2.3 kilobases. show that contains...
The expression of the human HSP70 gene is induced by a wide range physiological stresses, including exposure to heat shock and heavy metals, or under nonstress conditions, such as in response serum stimulation. We have previously demonstrated that either case regulated at primary level transcription. To determine whether transcription mediated through single multiple genetic elements, we dissected sequences upstream start site constructing chimeric genes retaining variable amounts 5'...
We have examined the expression of heat shock protein (hsp70) gene in human cells. The transcription hsp70 and accumulation cytoplasmic mRNA is induced by serum stimulation. Populations HeLa cells embryonic kidney (cell line 293) were starved. Upon stimulation, level transiently increases between 12 18 hr to a 10-fold higher level. increased levels can be accounted for 10- 15-fold increase rate gene. When serum-stimulated presence an inhibitor DNA synthesis, 1-beta-D-arabinofuranosylcytosine...
Bayesian inference provides a powerful approach to system identification and damage assessment for structures. The application of method is motivated by the fact that inverse problems in structural engineering, including health monitoring, are typically ill-conditioned ill-posed when using noisy incomplete data because various sources modeling uncertainties. One should not just search single “optimal” value vector model parameters but rather attempt describe whole family plausible based on...
Expression of the human gene encoding major heat shock protein, HSP70, was induced during cell growth by serum stimulation and after infection with adenovirus 5. In this study we showed that HSP70 expression could be 5 infection, even in absence exogenous factors. Whereas endogenous gene, it had no effect on early promoters. However, both cellular E3 promoter were activated infection. By using a collection reconstructed mutant viruses, identified 13S product E1A region as specific...
We have investigated the role of stacking faults in high quality ZnSxSe1−x heterostructures and a ZnSxSe1−x/CdxZn1−xSe based II-VI blue-green quantum well laser structure grown on GaAs substrates. find that these faults, which originate at epilayer/substrate interface during initial stages growth, act as sources for misfit dislocation formation region devices. analyzed mechanism dislocations. also show through cathodoluminescence microscopy nonradiative recombination centers therefore reduce...
The basal promoter of the human gene encoding 70-kDa heat shock protein (HSP70) controls maximal transcriptional activity and serum-regulated expression. We demonstrate that three elements defined by in vivo studies--CCAAT, element (SRE), "TATA"--correspond to protein-binding sites vitro. interactions with factors HeLa cell crude nuclear extracts were detected an exonuclease III digestion assay. sequence specificity was demonstrated probes containing wild-type sequences or unique...
Blue-green (λ=511 nm) separate confinement laser structures based on lattice-matched MgZnSSe-ZnSSe-CdZnSe have been grown by molecular beam epitaxy. Wide stripe gain-guided devices fabricated from several such wafers. These exhibit room-temperature pulsed threshold current densities as low 630 A/cm2 and voltages less than 9 V. Using a novel self-aligned process that results in planar surface, buried-ridge diodes also fabricated. demonstrated currents 2.5 mA, which is more factor of 50 lower...
The basal promoter of the human hsp70 gene is predominantly controlled by a CCAAT element at position -70 relative to transcriptional initiation site. We report isolation novel cDNA clone encoding 114-kDa polypeptide that binds promoter. Expression this CCAAT-binding factor (CBF) activated transcription from cotransfected promoter-reporter constructs in CCAAT-dependent manner. shows no homology previously identified or rat CCAAT/enhancer-binding protein.
Guided waves have been used for structural health monitoring to detect damage or defects in structures. However, guided wave signals often involve multiple modes and noise. Extracting meaningful information from the received signal becomes very challenging, especially when some of overlap. The aim this study is develop an effective way deal with noisy guided-wave detection as well de-noising. To achieve goal, a robust sparse Bayesian learning algorithm adopted. One many merits technique its...
A systematic dependence of the density and type stacking fault defects with substrate surface chemistry film growth mode was observed in ZnSe-based films grown on GaAs substrates. Namely, Frank-type faults is very large for Ga-rich surfaces, but low As-stabilized surfaces exposed to Zn prior film. In contrast, Shockley-type increases by 3D at initial stages growth, decreases greatly if are layer-by-layer mode. Films densities as 104/cm2 were obtained growing epilayers that 1–2 min films.
We have carried out a microstructural study of the 〈100〉 dark line defect that forms during degradation II-VI blue-green light emitters. find these defects lie in or near ZnCdSe quantum well and do not correspond to readily observable dislocation network transmission electron microscopy studies. speculate they may consist point small complexes. also estimates migration rates device operation can give rise such degradation.
We study the growth of pseudomorphic ZnSSe layers on GaAs. The dependence epilayer quality Zn exposure to GaAs surface is investigated. treatment prior ZnSe buffer As-rich results in lowest defect density. Transmission electron microscopy studies show that an atomic scale smooth interface formed. Based etch pit density and photoluminescence image analysis, with ≤1×104 cm−2 can be reproducibly obtained.
Expression of the human hsp70 gene is cell cycle regulated and inducible by both serum adenovirus E1a protein (K. Milarski R. Morimoto, Proc. Natl. Acad. Sci. USA 83:9517-9521, 1986; M. C. Simon, K. Kitchener, H.-T. Kao, E. Hickey, L. Weber, Voellmy, N. Heintz, J. Nevins, Mol. Cell. Biol. 7:2884-2890, 1987; B. Wu, H. Hurst, Jones, 6:2994-2999, Wu 82:6070-6074, 1985). Guided wave (GW) testing has become one of the most important nondestructive tools for evaluation structural in-service degradation. Although GW was widely used to inspect and screen many engineering structures, there are still challenges associated with its applications, which mostly originate from dispersive multi-mode nature signals as well noise contamination. To deal these challenges, an effective signal processing technique is introduced that enables accurately recover multiple modes...

We have carried out an electron-microscopy-based microstructural study of molecular beam epitaxially grown ZnxCd1−xS epilayers on GaAs(001) substrates. find that sulfur attacks bare GaAs substrates, resulting in pits act as nucleating centers for defects propagate into the epilayer. Such pitting may be avoided by capping substrate with a ZnSe buffer sulfur-free separate growth chamber. also investigate pseudomorphic zinc blende to wurtzite crystal structure change epitaxial and propose model...
