Extracellular vesicles (EVs) were isolated from Pectobacterium zantedeschiae culturing media using direct ultracentrifugation (UC), iodixanol cushion (ICUC), and density gradient (IDGUC) techniques. The isolates characterized with total protein content assay (bicinchoninic acid assay, BCA), nanoparticles tracking analysis (NTA), capillary electrophoresis (CE). A satisfactory correlation (R2 > 0.94) between quantitative results obtained BCA, NTA CE was achieved only for the IDGUC. proved...
Extracellular vesicles (EVs) are an experimental class of drug carriers. Alternative sources EVs currently being explored to overcome limitations related their manufacturing from mesenchymal stem cells. In this work, Citrus limon-derived were tested as carriers for the widely used chemotherapeutic – doxorubicin (DOX). Capillary electrophoresis (CE) and nanoplasmonic sensing (NPS) developed quality control DOX–EV preparations. It was found that CE method enables simultaneous detection free...
Bacteria of genus Pectobacterium are Gram-negative rods the family Pectobacteriaceae. They causative agent soft rot diseases crops and ornamental plants. However, their virulence mechanisms not yet fully elucidated. Membrane vesicles (MVs) universally released by bacteria believed to play an important role in pathogenicity survival environment. Our study investigates MVs Pectobacterium. The results indicate that morphology production depend on growth medium composition. In polygalacturonic...
Abstract The capillary zone electrophoresis (CZE) has recently been proposed by our group as a novel technique for outer membrane vesicles (OMVs) characterization ( J. Chromatography 1621 (2020) 461047 ). In present work the impact of selected parameters CZE method on OMVs isolates analysis was assessed. It shown that extension sample injection plug length significantly improves detectability macromolecular aggregates in CZE. Moreover, negligible adsorption to both uncoated and...
Despite the abundance of available cell lines, nearly 70% all recombinant therapeutic proteins today are produced in Chinese hamster ovary (CHO) cells. The impact protein overproduction on secretion exosomes by CHO cells has been investigated here. Increased extracellular vesicles (EVs) overexpressing was demonstrated with content assay, nanoparticle tracking analysis, and capillary electrophoresis. Our results revealed that a might induce EVs secretion, which be accompanied sequestration...
