Abstract The importance of diadinoxanthin (Ddx) de‐epoxidation in the short‐term modulation temperature effect on photosynthetic membranes diatom Phaeodactylum tricornutum was demonstrated by electron paramagnetic resonance (EPR), Laurdan fluorescence spectroscopy, and high‐performance liquid chromatography. 5‐SASL spin probe employed for EPR measurements provided information about membrane area close to polar head groups lipids, whereas with 16‐SASL probe, hydrophobic core, where fatty acid...
The effect of optimal and stress temperatures on the growth kinetics Phaeodactylum tricornutum CCAP/1055/1 strain (a model diatom with a known genome sequence) in batch cultures was examined. analysis obtained results showed two phases culture growth. There were significant positive correlations between OD increase chlorophyll c protein concentration at different temperatures. Fv/Fm parameter achieved maximum level 6(th) or 7(th) day then decreased to values registered first observation....
Zeaxanthin epoxidase (ZE) is an enzyme operating in the violaxanthin cycle, which involved photoprotective mechanisms. In this work model systems to study zeaxanthin (Zx) epoxidation were developed. Two assay are presented of Zx was observed. these assays two mutants Arabidopsis thaliana have active only one xanthophyll cycle enzymes used. The npq1 mutant possesses ZE and thus able convert (Vx) but de-epoxidase (VDE) inactive, so that Vx cannot be converted Zx. other mutant, npq2, VDE can...
To determine the role of α- and γ-tocopherol (TC), this study compared response to salt stress (200 mM NaCl) in wild type (WT) Arabidopsis thaliana (L.) Heynh. And its two mutants: (1) totally TC-deficient vte1; (2) vte4 accumulating γ-TC instead α-TC; (3) tmt transgenic line overaccumulating α-TC. Raman spectra revealed that salt-exposed α-TC plants were more flexible regulating chlorophyll, carotenoid polysaccharide levels than TC deficient mutants, while had lowest these biocompounds....
The greenhouse effect results in warming the planet's surface and a higher ocean heat content. It causes changes of water circulation frequency turbulent mixing, delivering warm water, phytoplankton nutrients CO2 from deeper layers to surface. All these can have an impact on Earth's ecosystems. In present study changing environmental temperature model diatom species, Thalassiosira pseudonana was tested under laboratory conditions. The purpose work analyze fluidity chemical composition thylakoid...
In the diadinoxanthin cycle epoxy group is removed from and diatoxanthin created. This conversion takes place e.g. in diatoms with involvement of enzyme de-epoxidase. one diatom species, Phaeodactylum tricornutum (CCAP 1055/1 strain genome sequenced) three de-epoxidase genes (PtVDE, PtVDL1, PtVDL2) have been identified, but only them (PtVDE) corresponds to violaxanthin de-epoxidase, an which commonly found higher plants. these studies, expression two another (UTEX 646), used were obtained...
During coal combustion soot formation is unwanted. There are many fuel additives used to reduce production. In this study toxicity and biodegradation of three types by two soil bacteria strains i.e. Pseudomonas aeruginosa Bacillus subtilis were compared. Two selected produce samples: (i) commonly containing significant quantities NaCl (40–44%) copper (Cu-additive), (ii) newly developed without with triiron tetraoxide instead (Feadditive). Average number bacterial colonies was similar when...
Using a pathogenic fungus and selected endophytic fungi of the ash tree, we propose modified model fungal dual cultures that allows us to obtain two new parameters particularly useful in search for pathogen control agents. The first these, called vitality parameter, is applicable study both biotic abiotic factors affecting growth. It measured as ratio growth radius direction studied factor opposite direction. second inhibition relates biofactors tested cultures. This parameter combines...
The purpose of this research was to obtain recombinant violaxanthin de-epoxidases (VDEs) from two species. first one VDE Arabidopsis thaliana (L.) Heynh. (WT Columbia strain) (AtVDE) which in vivo catalyzes conversion (Vx) zeaxanthin (Zx) via anteraxanthin (Ax). second Phaeodactylum tricornutum Bohlin, 1897 (CCAP 1055/1 (PtVDE) is responsible for de-epoxidation diadinoxanthin (Ddx) diatoxanthin (Dtx). As the step our experiments, open reading frames coding studied enzymes were amplified and...
