The successful design of nanofluidic devices for the manipulation biopolymers requires an understanding how predictions soft condensed matter physics scale with device dimensions. Here we present measurements DNA extended in nanochannels and show that below a critical width roughly twice persistence length there is crossover polymer physics.

ABSTRACT This article argues that ethnography is inadequately understood and recognized within administration science as a method for studying organizational culture. Ethnographic analyses of cultures are largely absent from the literature, primarily because such work derives social constructionist understanding science. The knowledge organizations thus provided interpretive, denying subject—object dichotomy inherent in mainstream empiricist applications analysis. In addition, whereas...

Abstract RAS-driven cancers comprise up to 30% of human cancers. RMC-6236 is a RAS(ON) multi-selective noncovalent inhibitor the active, GTP-bound state both mutant and wild-type variants canonical RAS isoforms with broad therapeutic potential for aforementioned unmet medical need. exhibited potent anticancer activity across RAS-addicted cell lines, particularly those harboring mutations at codon 12 KRAS. Notably, oral administration was tolerated in vivo drove profound tumor regressions...

Extensive fine-scale genetic diversity is found in many microbial species across varied environments, but for most, the evolutionary scenarios that generate observed variation remain unclear. Deep sequencing of a thermophilic cyanobacterial population and analysis statistics synonymous single-nucleotide polymorphisms revealed high rate homologous recombination departures from neutral drift consistent with effects hitchhiking. A sequenced isolate genome resembled an unlinked random mixture...

Microbial communities are commonly characterized by amplifying and sequencing target genes, but errors limit the precision of amplicon sequencing. We present DADA2, a software package that models corrects errors. DADA2 identified more real variants than other methods in Illumina-sequenced mock communities, some differing single nucleotide, while outputting fewer spurious sequences. analysis vaginal samples revealed diversity Lactobacillus crispatus strains undetected OTU methods.

PCR amplification and high-throughput sequencing theoretically enable the characterization of finest-scale diversity in natural microbial viral populations, but each these methods introduces random errors that are difficult to distinguish from genuine biological diversity. Several approaches have been proposed denoise data lack either speed or accuracy. We introduce a new denoising algorithm we call DADA (Divisive Amplicon Denoising Algorithm). Without training data, infers both sample...

The effects of intravenous naloxone on several the actions morphine (mean dose 30 mg/70 kg) were studied in six volunteer subjects. Naloxone produced a well defined reversal respiratory depression, analgesia, and miotic subjective morphine. agonist action outlasted antagonist single naloxone. effect repeated doses was also short-lived, but continuous infusions effective maintaining reversal.

Abstract Tumors acquire alterations in oncogenes and tumor suppressor genes an adaptive walk through the fitness landscape of tumorigenesis. However, interactions between that shape this remain poorly resolved cannot be revealed by human cancer genomics alone. Here, we use a multiplexed, autochthonous mouse platform to model quantify initiation growth more than one hundred genotypes lung tumors across four oncogenic contexts: KRAS G12D, G12C, BRAF V600E, EGFR L858R. We show is rugged—the...

Introduction The mesh choice for the majority of our retromuscular repairs is heavyweight knitted polypropylene (KP) mesh. However, supply chain issues necessitated a change to newer non-woven (NWP). We aimed evaluate initial experience with using NWP in abdominal wall reconstruction. Methods performed retrospective review all patients at institution who underwent elective, open incisional hernia repair or KP from January 2014 until December 2023. analyzed variables included patient...
