Intervertebral disc (IVD) degeneration (IVDD) is the main cause of low back pain with major social and economic burdens; however, its underlying molecular mechanisms remain poorly defined. Here we show that focal adhesion protein Kindlin-2 highly expressed in nucleus pulposus (NP), but not anulus fibrosus cartilaginous endplates, IVD tissues. Expression drastically decreased NP cells aged mice severe IVDD patients. Inducible deletion adult causes spontaneous striking IVDD-like phenotypes...

Low back pain impacts over 600 million people worldwide, predominantly due to intervertebral disc degeneration. This study focuses on the role of Piezo1, a crucial mechanosensitive ion channel protein, in pathology and potential treatment To investigate effects disc-specific Piezo1 deletion, we generated Aggrecan CreERT2 ; fl/fl mice examined both lumbar spine instability (LSI)- aging-induced Additionally, effect pharmacological inhibition was evaluated using GsMTx4, potent antagonist, an ex...

Osteoarthritis (OA) is the most common degenerative joint disease affecting older populations globally. Phosphatidylinositol-4-phosphate 5-kinase type-1 gamma (Pip5k1c), a lipid kinase catalyzing synthesis of phospholipid phosphatidylinositol 4,5-bisphosphate (PIP2), involved in various cellular processes, such as focal adhesion (FA) formation, cell migration, and signal transduction. However, whether Pip5k1c plays role pathogenesis OA remains unclear. Here we show that inducible deletion...

Abstract Degenerative disc disease (DDD) represents a significant global health challenge, yet its underlying molecular mechanisms remain elusive. This study aimed to investigate the role of type 1 phosphatidylinositol 4‐phosphate 5‐kinase (Pip5k1) in intervertebral (IVD) homeostasis and disease. All three Pip5k1 isoforms, namely Pip5k1α, Pip5k1β, Pip5k1γ, were detectable mouse human IVD tissues, with Pip5k1γ displaying highest expression nucleus pulposus (NP) cells. The was significantly...
